Objective:To investigate the effect of Naojian capsule on the expression of chemokine monocyte chemoattractant protein-1(MCP-1) and macrophage inflammatory protein-1α (MIP-1α) in rats after focal cerebral ischemia. Provide theoretical and experimental basis for clinical application for Naojian capsule.Methods:Using the method of middle cerebral artery occlusion (MCAO) to establish a permanent focal cerebral ischemia model.The rats were randomly divided into sham operation group, model group, BuYangHuanWu decoction group and Naojian capsule group,each group contains25rats. The rats were received different process after executed. Use arterial blood for hemorheology detection and enzyme-linked immunosorbent assay (ELISA) to detect MCP-1and MIP-1α content changes in serum. Use HE staining to observe the rats’ brain cells morphological distribution, and the immune histochemical method to detect the expression of MCP-1and MIP-1α content changes in rats’s brain.Results:1.The hippocampal neurons can appear typical ischemia tissue damage in rats after cerebral ischemia. The ischemia brain tissue could be improved recovery by the treatment of BuYangHuanWu decoction and Naojian capsules. Naojian capsule is working sooner thanBuYangHuanWu decoction.2. The plasma viscosity, whole blood high-shearing relative index, whole blood low-shearing relative index and Casson viscosity in hemorheology were all increased in rats after cerebral ischemia. The plasma viscosity, whole blood high-shearing relative index, whole blood low-shearing relative index and Casson viscosity in hemorheology could be reduced by by the treatment of BuYangHuanWu decoction and Naojian capsules, which compared with model group, the difference were statistically significant(P<0.05).3.The contain of MCP-1,MIP-1αin serum were significantly increased in rats after cerebral ischemia.The contain of MCP-1,MIP-1αcould be reduced by by the treatment of BuYangHuanWu decoction and Naojian capsules, which compared with model group, the difference were statistically significant(P<0.05).4.The contain of MCP-1,MIP-lain brain tissue were significantly increased in rats after cerebral ischemia.The contain of MCP-1,MIP-1αcould be reduced by by the treatment of BuYangHuanWu decoction and Naojian capsules, which compared with model group, the difference were statistically significant(P<0.05).Conclusion: 1.Naojian capsule can promote the tissue repair by improving the morphology distribution of hippocampal of ischemia brain tissue in rats after cerebral ischemia.2.Naojian capsule can improve the features of hemorheology and decrease the blood viscosity by reducing plasma viscosity, whole blood high-shearing relative index, whole blood low-shearing relative index and Casson viscosity.3.Naojian capsule can control the expression of chemokines in serum and brain tissue. It suggested that Naojian capsule may decrease the chemotaxis of inflammatory cells and inhibit the inflammatory response by reduce the expression of MCP-1, MIP-1α, which protect the brain.
