Backgrounds:Infrasound is a mechanical vibrational wave with frequency between 0.0001 ~20Hz, which human cannot hear. It has the same essence as ultrasound and audible sound and spreads in all kinds of elastic media (air, liquid, solid, etc.) in the form of alternating waves between rarefaction and compact form. It has the character of long effecting distance, strong penetration ability, and etc. Since Gavreau brought up the question about character and biologic effects of infrasound, with the development of science, especially of communication and industry, people began to know infrasound exists widely in our environment, and more and more countries and scientific institutions cognized importance to the damage caused by infrasound and carried out lots of research work. But until now there are little research and few reports about protection of infrasound. Traditional Chinese Medicine (T.C.M.) diagnoses and treats on the basis of an overall analysis of the illness and the patient's condition and combines signs and symptoms together in dialectical way and has its own advantage in treating disease. Former research had showed T.C.M. had certain effects in protecting tissue and organs such as heart and brain, in promoting immunity ang mitigating damages caused by free radicals.So we make use of T.C.M. to protect the damage caused by infrasound.Bu Yang Huan Wu Decoction, which was created by famous doctor Wang Qing-ren in Qing dynasty, first appeared in yi lin gai cuo It has the ability of invigorating qi and activating blood circulation. Bu Yang Huan Wu Decoction puts ingredients that invigorate qi and active blood circulation together as compatiblemedicines. Ingredients invigorating qi act as host, ingredients activating blood circulation act as supplement. Besides weakness of liver and kidney, weakness of qi and blocking of blood also appeared in the damage caused by infrasound, there is excessive rise of liver-yang. Except the upper function, white Hang chrysanthemum also has the ability of dredging feng and heat, clearing the eyes, calming the liver and subduing yang, clearing away heat and toxic material, so we add white Hang chrysanthemum to calm the liver and subdue yang, which we can provide overall protection from damages caused by infrasound.Objective:To study protecting effects against infrasound by Jia Wei Bu Yang Huan Wu Decoction,so as to provide theory for alleviating the damages caused by infrasound.Methods:BALB/c mice whose tearing and memory ability were close were randomized into normal control groups,infrasound control groups infrasound and drug-treated groups which were divide into preventive,therapeutic drug-treated groups based on different quomodo of drug-treated and divide into high,middle and low doses groups according to different dose of drug. Learing and memory ability, oxidation resistance in the brain (content of MDA, activity of GSH-Px and activity of SOD as yardsticks) and ultra-structure of pallium were observed after 14 days.Results: 1 Changes of learing and memory abilityAt 14th day after exposed to infrasound of 16Hz,130dB, mice in infrasound control group had longer escape time compared with normal control group, the difference had statistic means (P< 0.01) ; The groups that received medicine to therapy had less escape time compared with infrasound control group (P < 0.01), but there were little difference between different dosage groups (P>0.05) . There were little difference between mice in infrasound control group and preemptive medical treatment groups (P>0.05) , and there were little difference in groups with different dosage groups (P>0.05) .AT 14th day after exposed to infrasound of 8Hz,130dB, mices in infrasoundcontrol group had longer escape time compared with normal control group, the difference had statistic means (P<0.01) ;The groups that received medicine to therapy had less escape time compared with infrasound control group (P>0.05) andthere were little difference among groups received medicine with different dosage (P>0i05). There were little difference between mice in infrasound control group and preemptive medical treatment groups (P>0.05) , and there were little difference in groups with different dosage (P>0.05) .2 Changes of SOD activity, GSH-Px activity and content of MDA2.1 Groups of 16Hz, 130dBIn the groups that received medicine to therapy, activity of SOD in infrasound control group was higher compared with normal control group but without statistic difference (P>0.05 ) , and activity of SOD in every groups that received medicine to therapy was much higher than infrasound control group ( P < 0.05 ) , especially in high and middle dosage groups which had statistic difference compared with low dosage group (P< 0.05) ; Activity of GSH—Px in infrasound control group was higher compared with normal control group and had statistic means (P < 0.01) , and in every group that received medicine was much higher than in infrasound control group (P < 0.01) , especially in high dosage group which had statistic difference compared with in middle and low dosage groups (P < 0.01) ; Content of MDA in infrasound control group was higher compared with normal control group and had statistic means (P < 0.01) , and in every group that received medicine was lower than in infrasound control group (P< 0.05) , but there were little difference among different dosage group (P>0.05) .In the groups that received medicine in preemptive way, activity of SOD in infrasound control group had little difference compared with normal control group (P>0.05) , and in every group that received medicine had little difference compared with infrasound control group (P>0.05 ) , there were little difference among different dosage (P>0.05) ; Activity of GSH—Px in infrasound control group was higher compared with normal control group and had statistic means (P < 0.05) , activity of GSH—Px in low dosage group was much higher than infrasound control group (P <0.05) ,but there were little difference among high and middle dosage groups and infrasound control group (P>0.05) .Content of MDA in infrasound control group was higher compared with normal control group and had statistic means (P<0.01) , content of MDA in every group that received medicine was lower than infrasound control group but without statistic difference (P>0.05) ,and there were little difference among different dosage groups (P>0.05) . 2.2 Groups of 8Hz, 130dBIn the groups that received therapeutic medicine.activity of SOD in infrasound control group was lower compared with normal control group and had statistic means (P < 0.05) , and in every group that received medicine was much higher than in infrasound control group (P< 0.01) , especially in high dosage group which had statistic difference compared with in low dosage group (P < 0.05) ; Activity of GSH —Px in infrasound control group was higher compared with normal control group and had statistic means (P < 0.05) ,and in every group that received medicine was much higher than infrasound control group (P < 0.01) , especially in high dosage group but which had no statistic difference with middle and low dosage groups (P>0.05) ; Content of MDA in infrasound control group was higher compared with normal control group and had statistic means (P < 0.01) , and in high and middle dosage groups were lower than infrasound control group (P < 0.05) , but there were little difference between low dosage group and infrasound control group (P>0.05) .In the groups that received medicine in preemptive way, activity of SOD in infrasound control group was low compared with normal control group (P < 0.05) , and in groups that received medicine was higher than infrasound control group with statistic difference (P < 0.01) ,but there were little difference among different dosage groups (P>0.05) ; Activity of GSH—Px in infrasound control group was higher compared with normal control group and had statistic means (P < 0.05) ,and in different dosage groups were higher than infrasound control group but withoutstatistic difference (P>0.05) and there was no statistic difference among difference dosage groups (P>0.05) . Content of MDA in infrasound control group was higher compared with normal control group and had statistic means (P < 0.01) ,and in groups that received medicine was lower than infrasound control group but without statistic difference (P>0.05) ,and there were little difference among different dosage groups (P>0.05) .3 Changes of ultra-structure in palliumObserving pallium of mice in infrasound control group by transmission electron microscope, which exposed infrasound of 16Hz, 130dB after 14 days: The integrity of some neuron was damaged, glial cells appeared and chromatin apparently edged, heterochromatin appeared; mitochondrion was mild swelling and vacuolization, Golgi body was mild swelling; there were apparent gaps among capillary, medullary sheath was apparently separated and lots of lipofuscin appeared. After treated with the soup in therapeutic dosage, the damage became less, which mainly laid in that form of neuron was almost normal, heterochromatin decreased, swelling and vacuolization of mitochondrion became less, gaps among capillary became less and lipofuscin decreased, etc. the upper change was different according to different dosage. While in high, middle and low dosage groups which received medicine in preemptive way, ultra-structure in pallium was close to that of infrasound control group and damnification didn't lighten.Observing cortex of mice in infrasound control group by transmission electron microscope, which exposed infrasound of 16Hz, 130dB after 14 days: The form of neuron and was normal, swelling and hypertrophy of microglial appeared surrounding neuron. Mitochondrion was apparently deformed into fork or triangle and vacuolization was also observed; Golgi body was mild swelling; there were apparent gaps among capillary, medullary sheath became disordered and lots of lipofuscin appeared. After treated with the soup in therapeutic dosage, the damage became less, which mainly laid in that form of neuron and nuclei was almost normal, chromatin was mainly composed of euchromatin; swelling and vacuolization of mitochondrion became less, and Golgi body was less swelling; form of capillary became normal andlipofuscin became less. While in high, middle and low dosage groups which received medicine in preemptive way? ultra-structure in pallium was close to that of infrasound control group and damnification didn't lighten.Conclusion: Infrasound can bring damages to mice, which mainly lies in decline of learning and memory ability, changes of activity of SOD and GSH-Px,content of MDA in the body increaseing,the change of ultra-structure in palliumJia Wei Bu Yang Huan Wu Decoction treated by therapy can improve learning and memory ability by promoting the activity of SOD and GSH—Px, eliminating excessive free radicals, lowering lipid per oxidation to decrease the content of MDA and lighten the damnification of pallium .Different dose has different protection effects. But preventive drug-treated groups'affects were not evidence.
