Leptin is a protein hormone produced by fat cells. The expression of the gene is named the OB gene. The main function of leptin is to control weight. The mutation of the OB gene leads to the increase of mice’s body fat, the increase of diet, and the change of relative factors. The main part in which leptin functions is the thalamus, its way to control the body’s hormone secretion is through the hypothalamus neuromodulation, to achieve the effect of reducing body fat in all aspects. Obesity in the modern society has become a quite common phenomenon. the discovery of leptin helps us know more about the mechanism of and causes for obesity. However, leptin failed to help the obese to lose weight. A large number of experiments found that injections of leptin on weight loss is not ideal. Researches on the relationship between leptin and movement mainly focus on whether sports exercise can cause changes of leptin in the body. However, there is no research about the influence of leptin or the influence of movement ability on some indicators of organism movement. The experiment is based on the combination of exercise, obesity, and leptin injections, aims to analyze the effects of leptin on movement through the combination of the three, the effects of leptin on obesity, or the effects of a combination of the three or the effect of the three respectively on mice.Research methods:Choose32mice from the36SD mice (male,sterile level) through obesity modeling. Divide them into four groups, namely obesity quiet control group (O group), obesity movement control group (OSgroup), obesity and leptin quiet group (OL group), and the obesity movement leptin group (OSL group). OS group and OSL group conduct swim training with fixed timing duration of31days, and eight days before the end of the trial, all mice take an exhaustive test. Once a day OL group and OSL group have leptin intravenous the injection with the volume of0.05ml for the weight of100g of mice. The standard is150ml solution configuration with0.6mg rat leptin protein thin prime solution. record the weight changes of mice, and take the blood, soleus muscle tissue, and liver tissue at the end of the experiment; in order to test the soleus muscle wet weight, the detection of biochemical markers the rat hindlimb soleus muscle and liver tissue in Ca2+-ATPase activity of Na+K+-ATPase activity of succinate dehydrogenase (SDH) activity, glycogen concentration in skeletal muscle mitochondria LPO content, hemoglobin and red blood cell count determination.Experimental results:(1) Na+K+-ATPase activity:the rat muscle organization OS group is larger than the group O Na+k+-ATPase activity (P<0.05), the OS group is larger than the OL group, OLS group (P <0.01). The remaining three groups of Na+K+-ATPase activity was no significant difference (P>0.05). Liver tissues OS group larger than the OL group (P<0.05), and the other group comparison are no significant differences (P>0.05).(2) Ca2+-ATPase activity:OL group is smaller than the O group, the OS group (P<0.05), other groups had no significant difference (P>0.05). There was no significant difference (P>0.05) in the liver.(3)Rats muscle SDH activity:OS group were larger than the other three groups (P<0.05), compared to the other groups, there was no difference (P>0.05). Liver SDH activity there was no significant difference (P>0.05).(4)The muscle mitochondrial lpo content:OS is less than the OSL group (P<0.05), and other groups shows no significant difference (P>0.05).(5)Muscle glycogen content:OSL larger than the OS group (P<0.05), and the other four groups of data showed no significant difference (P>0.05). The liver four sets of data showed no significant difference (P>0.05).(6)The number of red blood cells and rat hemoglobin content:OSL group is larger than the O group (P<0.05), and other groups, there was no significant difference (P>0.05).(7)Soleus wet weight:OS group than in the O group OL group (P>0.05), the rest of the group was no significant difference (P>0.05).(8)The rats rate of weight change:O group larger than OS group, OL groups, OSL group (P<0.05). OSL group less than O group OS groups, OL group (P<0.05), and the other group showed no significant difference (P>0.05).(9)After exhaustive exercise a few days of rats weight, diet have changed a lot, the OL group decreased more obvious than other groups. Conclusions:(1) Exercise with leptin injections are to reduce the growth rate of rats weight, a combi-nation of both can play better the effect of lowering the rate of weight gain.(2) Through the exercise,can increase the rat hindlimb soleus muscle tissue’s Na+K+-ATPase activity, Ca2+-ATPase activity, and the SDH activity, and can significantly improve hindlimb halibut muscle wet weight.mean its a good improvement of movement ability.(3)The quiet mice with leptin injections will lead to reduced muscle Ca2+-ATPase activity, tis bad to athletic ability.(4) The exercise rats leptin injection will result in the the rat hindlimb soleus Organi-zation Na+K+-ATPase activity, succinate dehydrogenase (SDH) activity decreased, as well as lead to rats in the soleus muscle mitochondria of LPO concentration increased, the soleus muscle wet weight and not as a the OS group increased significantly, and therefore be adverse effects related to the movment ability of the rats.(5) Exhaustive movment later, body weight in obese rats, the next day a more significant impact. Rats, especially injection of leptin still evident.(6) OSL group of muscle glycogen content is larger than the OS group (P<0.05), but the liver glycogen compared among the groups, there was no difference (P>0.05), mean the OSL group exercise capacity is higher thanOS group.
