Bamboo is a perennial evergreen plant, many parts of it are beneficial to the human body, Thebamboo shoots have nourish yin, appetizers, heat, phlegm and other effects. Bencao Gangmu described in the bamboo leaves have Chufan heat convulsion effect.Now,there are also scholars reported thatflavonoids in bamboohave the effect of drug food, antibacterial, anti-virus activity and so on, is a rare natural medicinal material.This paper selects Bambusa surrecta stem as raw material, first from the bamboo stem in the presence of flavonoid compounds, extraction after impurity removal, of flavonoids species, antibacterial effect were analyzed study, first from the bamboo stem in the presence of flavonoid compounds, extraction after impurity removal, of flavonoids species, antibacterial effect were analyzed study.The main research results are showen below.Currently, in modern society, the vast majority of food preservatives are synthesized through chemical method, improperly using can cause serious damage to human, therefore, looking for a safe and effective natural preservatives is the current focus in the field of preservatives. Bamboo flavonoids is a kind of natural active ingredients. In recent years, there have been studies suggest that flavonoids extracted from bamboo leaves have fight free radicals, anti-oxidation, fall hematic fat, resisting bacteria and other pharmacological effects.This article selects the stem of Bambusa surrecta as material,and it is the first time to extract flavonoids from the stem of Bambusa surrecta,optimize the extr-action process, separate and purify the production through macroporous resin adsorption method, at the same time, invest- ation on the bacteriostatic activity of flavonoids from bamboo stem, provides reference for the application of future.The main research results are as follows:Using the organic solvent though hot reflux method to extract the flavonoids from stem of bamboo, investigating single factor’s influence on the extraction yield such as type and concentration of extractant, liquid ratio, extraction temperature、method and time and then deeply optimized by using the orthogonal experiment method for the extraction process. The results show that the extra-cting agent concentration had the greatest influence on extraction yield, the influence of material liquid is the second, minimum the influence of extraction temperature, at the same time, it is concluded that the best extraction process is: 50% ethanol aqueous solution as the extraction agent, hot reflux extraction 2 h under 80 ℃, and solid-liquid ratio of 1:20(g/mL), under the condition, the stem of bamboo flavonoids extraction yield is 1.86%.Isolation and purification of crude flavonoids extracted from bamboo stem though the macroporous resin adsorption method, compared with the behaviors of the seven kinds of macroporous resin a-dsorption rate and desorption rate, the type AB-8 macroporous resin is ideal candidate. Using type AB-8 macroporous resin to separateion and purification of extract, the optimal parameters as follows:1.0 mg/mL flavonoids, with pH=6, at the rate of 1.0 mL/min and the sample volume is 170 mL.The products were washed with120 mL ethanol elution(70%) on the rate of 1.5 mL/min. This is the best condition, the quality of flavonoids from stem bamboo increses from 21.3% to 50.85%, 2.4 times as before.The element of product was characterized by HPLC, though setting of chromatographic conditions and mobile phase,have successful separate and determinate rutin, rhizoma kaempferiae respectively glycosides, st John’s wort glycosides, rhizoma kaempferiae glucuronic acid glycosides, rhizoma kaempferiae phenol 5 element of flavones from stem bamboo.Final test effect of by plate count method and the bacteriostatic circle method, containing staphylococcus aureus, escherichia coli, bacillus, rhizopus, mucor. The results show that st-em bamboo flavonoids have actively antibacterial to bacteria and mold,under certain concentration,for the more function with bacteria have a better result than mold.The value of PH has great influence to the bacteriostatic activity. In acidic environment, bamboo stem flavonoids bacteriostatic effect is better, at the same time, bamboo flavonoids had good thermal stability, heat treatment at 100 ℃ bacteriostasis is still good,over 120 ℃, the bacteriostatic activity decrease. We can make a conclution that stem bamboo flavonoids is a kind of natural preservatives can resistance to high temperature.
