ZD preparation was made based oil many years of research and clinical application of prescription, which was a composite Chinese medicine preparation applied to treat the injured soft tissue of horses’hooves, made into a modern type of topical ointment medicine combining various excipients. To test the toxicity, the detection of drug toxicity in mice was measured by median lethal dose, the maximal tolerance dose and skin irritation test. Through the mice and horses soft tissue damage modeling and application of ZD preparations for treatment, by radioimmunoassay to detect the dynamic changes in IL-1β, IL-6, IL-8and IL-10secretion, exploring the impact of inflammatory cytokines so as to ZD preparations for treatment equine animal soft tissue injury mechanisms provide the basis.Our experimental results indicated that:(1) By gavage in mice there was no median lethal dose of drugs detected in this aqueous extract, it was safe for the maximal tolerance dose of the drug into the body and has little toxicity effects. There was no significant differences of ZD preparation shown on the skin irritation tests in experimental and control groups of mice, proving application of this preparation was less toxic, safe and reliable for topical use.(2) In the experiments of ZD preparations for the treatment of modeling in mice, the secretion of IL-1β, IL-6and IL-8were increased in the first day, while IL-10exhibited a low level. With the drug treatment of inflammatory response, IL-1β, IL-6and IL-8levels were decreased while the level of IL-10was increased. ZD preparation of high dose therapeutic effect was similar to positive drug treatment groups.(3) The experiment on equine model which soft tissue was injured showed the site of injury swelling significantly, basic physiological indicators did not change, cells associated with inflammation obviously changed in blood, there was also significant changes in pathological examination. After ZD preparations treatment, the trend of changes in serum inflammatory cytokines (IL-1β, IL-6,IL-8and IL-10) were consistent with the results of mice model.
