The Influnence Of ZD Preparation On Repairs Of Soft Tissue Injury And Macrophage Cell Line RAW264.7

ZD preparation for external use, developed in accordance with senior professors’ proved recipe, is a pure traditional Chinese medicine preparation applied to treat the injured soft tissue of horses’hooves. This prescription consists of frankincense, myrrh, angelica, safflower and other 12 kinds of traditional Chinese medicine. Our experimental results by preliminary experiment indicated that:ZD preparation has good effects for treating the injured soft tissue and has therapeutic, Anti-inflammatory, analgesia, promoting the role of swelling.ZD preparation was used for clinical application in the treatment of soft tissue injury of hose has a good therapeutic effect. In order to observe the anti-inflammatory effect of ZD preparation. A model of acute soft tissue injury in mice and the inflammatory model of macrophages were established.Healthy Kunming mice were randomly divided into normal group, model group, positive drug group and experimental drug group. In addition to the normal group, the other groups were prepared with soft tissue injury model, the pathological changes of mice were observed to determine whether the model was successful. After modeling 1 day began to give the patient a smear drug,2 times a day, to the drug 7th day. At the same time point after 1 st day and 7th day in the same time each group of 6 mice were randomly assigned to combat site as the center, to carry out muscle, and the expression of IL-1 beta and IL-6 in mouse muscle was detected by immuno histochemistry.RAW264.7 macrophages line in the mice was induced by LPS to set up the inflammatory model. NO production was examined by Griess reaction, and the productions of IL-1 beta, IL-6 and TNF-alpha were assayed by enzyme-linked immunosorbent assay.After the mice model, when the muscle tissue have congestion edema and other symptoms, the mouse model is successfully established. After drug intervention, the symptoms and pathology of each drug group were improved in different degree. The expression levels of IL-1 beta and IL-6 in the muscle tissue of mice were significantly reduced compared with the model group.ZD preparations were used to do the prognosis, the supernatant of NO, TNF-alpha, IL-1 beta, IL-6 content decreased compared with the model group, which exhibited the inhibitory effect on LPS-induced the productions of NO, TNF-alpha, IL-1 beta, IL-6 in a dose-dependent and time-dependent manner.In summary, we studied the Soft tissue injury model in vivo and the anti-inflammatory mechanism of ZD preparations in vitro through the establishment of inflammation model of RAW264.7 macrophages induced by LPS via using IHC Griess and ELISA. Results show that ZD preparations could achieve its anti-inflammatory effects by inhibiting the expression of pro-inflammatory cytokines.
