The Impacts Of Shao’s Method Of Wu Zhen On The Expression Of A-SMA And FN Of Asthma Model Rats

ObjectiveThis topic through acupuncture at FengShu（BL13），DaZhui（DU14），FengMen（BL12）in asthma model rats. Observation of acupuncture on groupsof asthma model in rat lung tissue and serum a-SMA、FN expression and theinfluence of the influence of HE staining lung tissue pathology. To furtherexplore the mechanism of acupuncture and moxibustion treatment of asthmaand acupuncture mechanism of action of airway remodeling of asthma modelrats. And provide scientific theory basis for acupuncture and moxibustiontreatment of asthma.Method40grade clean heaithy male SD rats （which weight range from20to140g）were randomly divided into blank control group，model control group，drug treatment group and acupuncture treatment group，each group consisted often. By intraperitoneal injection of OVA solution and atomization inhalation ofthe1%of the OVA solution sensitization to establish asthma model（1times aday， atotal of7times）. Acupuncture treatment group acupuncture asthma ratsFengShu（double），DaZhui and FengMen（double）after daily stimulate. Drugtreatment group intraperitoneal injection0.05%dexamethasone sodiumphosphate solution after daily stimulate. Model control group don’t do specialtreatment and model control group intraperitoneal injection physiological salineinstead of OVA solution after daily stimulate. Within the last6hours aftertreatment of chloral hydrate anesthesia，we take off cervical executed aftereyeball blood in rats. Then take right lower lung tissues and embed inparaffin.Finally，emply the method of immunohistochemical to determine the expression of a-SMA and FN expression in lung tissue. To observation ratairway morphology change and a-SMA、FN in asthma model expression in ratlung tissue and serum.Results1. Groups of behavioral signs observed in rats：Rats model control group compared with blank control group， appearsneezing， choking cough， runny nose， wheezing， shortness of breath，depressed， and slow to respond. Blank control group rats agile action and noexception. Drug treatment group and acupuncture treatment group rats shoutssneezing， coughing， wheezing and other symptoms compared with modelcontrol group significantly decreased， and responsive.2. Each rat lung tissue morphology observation：Model control group rats with airway epithelial damage loss， the mucosaand submucosa and hyperplasia of smooth muscle layer， deformation；Airwaywall thickening and luminal narrowing of the lumen within a large number ofdischarge； Pulmonary interstitial inflammatory cell infiltration in greatquantities， alveolar wall cracking deformation， alveolar interval structuredisorder；Blood vessels， blood vessel wall thickening. But drug treatmentgroup and acupuncture treatment group have basic complete airway structureand less pulmonary interstitial inflammatory cell infiltration， essentiallynormal lung tissue structure. And blank control group rats with airway structureis complete， no obvious loss and distortion， did not see pulmonary interstitialinflammatory cell infiltration， morphological structure of normal lung tissue.3. Group a-SMA comparison of lung tissue in rats：Rare cytoplasm in blank control group rats lung tissue is tan dyed a-SMApositive cells. Model control group positive expression in lung tissue of ratsobviously increased， compared with the blank control group， the differenceis statistically significant (P<0.01).Acupuncture treatment group and drugtreatment group compared with model control group， positive expression inlung tissue decreased significantly， the difference is statistically significant(P<0.01)，but it is still higher than the blank control group. Acupuncturetreatment group compared with the drug therapy group， the difference isstatistically significant (P<0.01). 4. Group FN comparison of lung tissue in rats：Rare cytoplasm in blank control group rats lung tissue is tan dyed FNpositive cells. Model control group positive expression in lung tissue of ratsobviously increased， compared with the blank control group， the differenceis statistically significant (P<0.01).Acupuncture treatment group and drugtreatment group compared with model control group， positive expression inlung tissue decreased significantly， the difference is statistically significant(P<0.01)，but it is still higher than the blank control group. Acupuncturetreatment group compared with the drug therapy group， the difference isstatistically significant (P<0.01).5. Group a-SMA comparison of serum in rats：Model control group compared with blank control group， serum of rats ina-SMA concentration significantly increased， the difference is statisticallysignificant (P<0.01). Acupuncture treatment group and drug treatment groupcompared with model control group，a-SMA significantly lower concentration，the difference is statistically significant (P<0.01)，but it is still higher than theblank control group.Acupuncture treatment group compared with the drugtherapy group， the difference is statistically significant (P<0.01).6. Group FN comparison of serum in rats：Model control group compared with blank control group， serum of rats inFN concentration significantly increased， the difference is statisticallysignificant (P<0.01). Acupuncture treatment group and drug treatment groupcompared with model control group， FN significantly lower concentration，thedifference is statistically significant (P<0.01)，but it is still higher than theblank control group.Acupuncture treatment group compared with the drugtherapy group， the difference is statistically significant (P<0.01).Conclusion1. Shao’s Method of Wu Zhen can improve the symptoms of asthma modelrats.2. Shao’s Method of Wu Zhen can reduce lung tissue morphology changesof asthma model rats.3. Shao’s Method of Wu Zhen can reduce a-SMA and FN level Model inlung of asthma model rats. 4. Shao’s Method of Wu Zhen can reduce a-SMA and FN level Model inserum of asthma model rats.Shao’s Method of Wu Zhen can significantly reduce asthma model ratsa-SMA and the concentration of the FN content in lung tissue and serum，prevent airway remodeling， acupuncture reduce a-SMA， positive expressionof FN is one of the ways to asthma treatment.
