Objective: The purpose of this study was to observe the effections of Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique on motor function and the expression of Nogo A and Rho A in rabbits with sciatic nerve injury,and provide theoretical basis for the treatment of peripheral nerve injury by Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique.Methods: 180 New Zealand rabbits were randomly divided into one in 5groups: a normal control group,a nerve crush control group,three treatment groups(a Neurodynamic Mobilization Technique group,a Jiaji electroacupuncture group,and a Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique group),36 in each group.Followed by postoperative treatment time for 1 week,2 weeks,4 weeks and then divided into 3 subgroups,12 in each subgroup.Sunderland ? degree injury model of the sciatic nerve was made by clamping.The normal control group did not make any intervention.The nerve crush control group was placed in the cage quietly after surgery.The Neurodynamic Mobilization Technique group was treated by Neurodynamic Mobilization Technique.Each operation time was 1s,relax 5s.Do one set of 10 repetitions.6 days a week.The Jiaji electroacupuncture group was treated with Jiaji Electroacupuncture.The Jiaji Electroacupuncture group received a sterile needle inserted into Jiaji point from the sciatic nerve for Jiaji electroacupuncture,once/d,30 min/time,6times/week.The Jiaji electroacupuncture combined with neurodynamics group received Jiaji electroacupuncture first and then Neurodynamic Mobilization Technique.The function of injured sciatic nerve was evaluated by the toe-spreading index and Modified Tarlov scale;The sciatic nerve and the connected spinal cord were harvested from animals.RT-PCR method was used to observe the gene expression of Rho A changes;Western Blot method was used to observe the expression of protein factor Rho A changes.Since the expression of Nogo A is only in the central nervous system,the gene expression of Nogo A was detected by RT-PCR,and the expression of Nogo A protein was detected by Western Blot only in the connected spinal cord of sciatic nerve.Result:1.The toe-spreading index and modified Tarlov scale: At 1 week,2 weeks and 4 weeks,the scores of the nerve crush control group,three treatment groups were significantly reduced compared to that of the normal control group(P<0.05).The scores were improved in the three treatment groups compared to the nerve crush control group(P<0.05).Furthermore,the Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique group was higher than the Neurodynamic Mobilization Technique group,Jiaji electroacupuncture group,(P<0.05),and the groups treated for 4 weeks had better recovery(P<0.05).2.The expression of Nogo A m RNA and protein At 1 week,2 weeks and 4 weeks,the expression of Nogo A in the nerve crush control group,three treatment groups were higher than in the normal control group(P<0.05).The treatment groups had lower levels than the nerve crush control group(P<0.05).In addition,the Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique group had a lower level than the Neurodynamic Mobilization Technique group and Jiaji electroacupuncture group at the three given points in time(P<0.05).3.The expression of Rho A m RNA and protein(1)the connected L4-L6 spinal cord: At 1 week,2 weeks and 4 weeks,the expression of Rho A in the nerve crush control group,three treatment groups were higher than in the normal control group(P<0.05).The treatment groups had lower levels than the nerve crush control group(P<0.05).In addition,the Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique group had a lower level than the Neurodynamic Mobilization Technique group and Jiaji electroacupuncture group at the three given points in time(P<0.05).(2)Sciatic nerve:At 1 week,2 weeks and 4 weeks,the expression of Rho A in the nerve crush control group,three treatment groups were higher than the normal control group(P<0.01),and three treatment groups were lower than the nerve crush control group(P<0.01).At 2 weeks and 4 weeks,the Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique group had significantly lower Rho A m RNA expression than the Neurodynamic Mobilization Technique or Jiaji electroacupuncture group(P<0.05).At 1 week,the Jiaji electroacupuncture group had lower expression than the Neurodynamic Mobilization Technique group or the Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique group(P<0.05),but there was no significant difference between the Neurodynamic Mobilization Technique group and the Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique group(P>0.05).Conclusions: Neurodynamic Mobilization Technique and Jiaji electroacupuncture can promote the repair of injured sciatic nerve,which may be related to the decrease of Nogo A and Rho A expression;Jiaji electroacupuncture combined with Neurodynamic Mobilization Technique was more effective than either intervention alone.
