Objective: To observe the effect of Jiaji electroacupuncture combined with Nerve mobilization on the number of myelinated axons and the expression of netrin-1 and Slit2 protein in the injured nerve and spinal ganglion after sciatic nerve injury in rabbits,and to study the mechanism of Jiaji electroacupuncture combined with Nerve mobilization in the process of axonal regeneration,so as to provide practical and theoretical support for the clinical research and treatment of peripheral nerve injury.Methods: 75 New Zealand rabbits were randomly separated into five groups: tow control groups: normal group and model group;three treatment groups : Nerve mobilization group,Jiaji electroacupuncture group and Jiaji electroacupuncture combined with Nerve mobilization group,15 rabbits in each group.According to the treatment time,the rabits was divided into three subgroups: one week,two weeks and four weeks.Normal control group: grasp on time without any other treatment.Model control group: after the model was established,the rabits were fed in a quiet and sanitary environment.Nerve mobilization group: the rabits were treated with Nerve mobilization.Jiaji electroacupuncture group: treated with Jiaji electroacupuncture.Jiaji electroacupuncture combined with Nerve mobilization group: the rabits were treated with Jiaji electroacupuncture combined with Nerve mobilization.After the treatment of each group,the samples were taken at the time point,the number of myelinated axons in the sciatic nerve and spinal cord was detected by toluidine blue staining,and the expression of netrin-1 and Slit2 protein was detected by immunohistochemistry.Result:(1)The number of myelinated axons: the number of myelinated axons of spinal cord and sciatic nerve in the three treatment groups(Nerve mobilization group,Jiaji electroacupuncture group,Jiaji electroacupuncture node combined with Nerve mobilization group)was higher than that in the model group(P< 0.05),and that in the three treatment groups was higher thanthat in the other two groups(P < 0.05).(2)Expression of Netrin-1 protein: the expression of netrin-1 proteinof spinal cord and sciatic nerve in the model group and the treatment group was up-regulated compared with that in the normal group at 1 and 2 week time points,down regulated at 4 week time points,lower than that in the normal group,and higher than that in the model group at 3 time points(P < 0.05),in which the expression in the electroacupuncture combined with Nerve mobilization group was higher than that in the other two treatment groups(P<0.05).(3)Expression of Slit2 protein: the expression of Slit2 protein in the model group and the treatment group of spinal cord and sciatic nerve was higher than that in the normal group at one week and two week time points,and decreased at four week time points,lower than that in the normal group.The expression of netrin-1 protein in the three treatment groups at three time points was higher than that in the model group(P<0.05).The expression of netrin-1 protein in the Jiaji electroacupuncture combined with Nerve mobilization group was higher than that in the other two treatment groups.(P<0.05).Conclusion:(1)Jiaji electroacupuncture,Nerve mobilization and Jiaji electroacupuncture combined with Nerve mobilization can increase the number of myelinated axons of sciatic nerve,up regulate the expression of netrin-1 and Slit2 protein,so as to promote the regeneration of sciatic nerve after injury.The combination effect of the two is better.(2)Jiaji electroacupuncture combined with Nerve mobilization can promote axonal regeneration after sciatic nerve injury,which may be related to up regulating the expression of netrin-1 and Slit2 protein in injured nerve and its corresponding spinal cord segments.
