| OBJECTIVE: By observing the clip ridge electric acupuncture combined with neural mobilization to rabbit sciatic nerve conformity of lower limb movement function after injury,and the Ras-related C3 botoxin substrate1 gene and protein substrates,F-actin expression,the influence of the clip ridge electric acupuncture combined with neural mobilization to hemiresected lumbar rabbit sciatic nerve regulation mechanism,to clip ridge electric acupuncture combined with neural mobilization provides the theory basis for treatment of peripheral nerve injury.METHODS: 180 New Zealand rabbits were randomly divided into normal control group,model control group,Jiaji electroacupuncture group,nerve mobilization group,and Jiaji electroacupuncture combined with nerve mobilization group.Thirty-six rabbits in each group were divided into three subgroups after 1 week,2 weeks,and 4 weeks of treatment,and 12 New Zealand rabbits in each subgroup.In the model control group,Jiaji electroacupuncture group,nerve mobilization group,Jiaji electroacupuncture combined with nerve mobilization group,the sciatic nerve injury model was induced by clamping method.The normal control group and the model control group did not have any intervention.The nerve mobilization group was treated with nerve mobilization,the Jiaji electroacupuncture group was treated with Jiaji electroacupuncture,and the Jiaji electroacupuncture combined with nerve mobilization group was treated with Jiaji electroacupuncture and nerve mobilization.After 1,2,and 4 weeks of treatment,the toe reflex score and modified Tarlov score were used to assess the functional recovery of the lower extremities in 5 groups of New Zealand rabbits.After 1,2,and 4 weeks of treatment,12 New Zealand rabbits in each group were sacrificed by exsanguination and the sciatic nerve and corresponding segments of the spinal cord(L4-L6 segments)were harvested.The expression of Ras-related C3 botoxin substrate 1 mRNA and protein was detected by polymerase chain reaction(PCR)and Western blot.RESULTS: After 1,2 and 4 weeks of treatment,the sciatic nerve function of the New Zealand rabbits in Jiaji electroacupuncture group,nerve mobilization group and Jiaji electroacupuncture combined with nerve mobilization group were higher than those in the model control group at the same time point,and the differences were statistically significant.Significance(P<0.05),but lower than the normal control group at the same time point,the difference was statistically significant(P<0.01);and Jiaji electroacupuncture combined with nerve mobilization group at each time point The sciatic nerve function was higher than that of Jiaji electroacupuncture group and nerve mobilization group at the same time point,and the difference was statistically significant(P<0.05).After 1,2 and 4 weeks of treatment,the expression of Ras-related C3 botoxin substrate 1 mRNA in spinal cord and sciatic nerve of Jiaji electroacupuncture group,nerve mobilization group and Jiaji electroacupuncture combined with nerve mobilization group was higher than that of the model control group at the same time point,but lower than that of the normal control group at the same time point(P<0.01),and the expression of Ras-related C3 botoxin substrate 1mRNA in spinal cord and sciatic nerve of Jiaji electroacupuncture combined with nerve mobilization group was higher than that of Jiaji electroacupuncture group and nerve mobilization group at the same time point(P<0.01).After 1,2,and 4 weeks of treatment,the expression of Ras-related C3 botoxin substrate 1 protein in the spinal cord of Jiaji electroacupuncture group,nerve mobilization group,and Jiaji electroacupuncture combined with nerve mobilization group was higher than that of the model control group at the same time point,but lower than that of the normal control group at the same time point,and the difference was statistically significant(P<0.01).After 1 week of treatment,the expression of Ras-related C3 botoxin substrate 1 protein in the spinal cord of Jiaji electroacupuncture combined with nerve mobilization group was significantly higher than that of Jiaji electroacupuncture group(P<0.01).After 2 and 4 weeks of treatment,theexpression of Ras-related C3 botoxin substrate 1 protein in the spinal cord of Jiaji electroacupuncture combined with nerve mobilization group was significantly higher than that of Jiaji electroacupuncture group and nerve mobilization group at the same time point,and the difference was statistically significant(P<0.01).After 1 week of treatment,the expression of Ras-related C3 botoxin substrate 1 protein in the sciatic nerve of Jiaji electroacupuncture group,nerve mobilization group,and Jiaji electroacupuncture combined with nerve mobilization group was lower than that of the model control group and the normal control group,and the difference was statistically significant(P<0.01).After 2 and 4 weeks of treatment,the expression of Ras-related C3 botoxin substrate 1 protein in the sciatic nerve of Jiaji electroacupuncture group,nerve mobilization group,and Jiaji electroacupuncture combined with nerve mobilization group was higher than that of the model control group at the same time point,which was lower than that of the normal control group at the same time point(P<0.01).The expression of Ras-related C3 botoxin substrate 1 protein in the sciatic nerve of Jiaji electroacupuncture combined with nerve mobilization group was significantly higher than that of Jiaji electroacupuncture group and nerve mobilization group at the same time point.The differences were statistically significant(P<0.01).By immunofluorescence protein staining analysis,after 1,2,and 4 weeks of treatment,the expression of F-actin protein in the spinal cord of Jiaji electroacupuncture group,nerve mobilization group,and Jiaji electroacupuncture combined with nerve mobilization group was higher than that of the model control group and the normal control group at the same time point,the difference was statistically significant(P<0.01).CONCLUSION: Neurolysis combined with Jiaji electroacupuncture can promote the regeneration of axons after sciatic nerve injury.The mechanism may be related to the up-regulation of Ras-related C3 botoxin substrate 1gene,F-actin and protein expression in injured sciatic nerve and corresponding spinal cord segments. |
