Purpose: Through the observation of Huoxue Xifeng method for migraine onmonoamine neurotransmitters in the brain tissue pathological changes, the preventive andtreatment after the behavior of rats5-hydroxytryptamine(5-HT), dihydroxyphenylethylamine(DA) and noradrenaline(NE) affect the content, and to explore the Huoxue Xifengmethod mechanism of preventive treatment of migraine.Method:1. Grouping:84Wistar rats, male and female were randomly divided into7groups: blank group, model group, Zhen Nao Ning group, sibelium group, Huoxue Xifengparticles of low, middle and high dose groups.2. Molding: continuous administration for7days, in the1h after the last administrationbegan preparing the animal model of migraine, the blank group were given subcutaneousinjection of10mg/kg saline, other groups in accordance with the subcutaneous injection of10mg/kg Nitroglycerin Injection.3. Ethology determination: using digital video recorders location shooting form, numberfrom molding start method to segment duration count was observed in rat general state andafter different time climbing cage, scratching his head. Is divided into4periods: subcutaneousNitroglycerin Injection after0~30min,31~60min,61~90min,91min~120min.4. Determination of the content of5-HT, DA, NE: behavioral observation, kill theexperimental rats, using enzyme-linked immunosorbent assay (enzyme linked immunosorbentassay, ELISA) in the brain tissue of rats in each group were detected in5-HT, DA, NE, toobserve the effects of Huoxue Xifeng method for migraine with blood stasis monoamineneurotransmitter contents in the wind rat brain.Result:1. Effects of Huoxue Xifeng method on the behavior of migraine rats: comparedwith the model group, Huoxue Xifeng particle dose, high dose group, sibelium group ratsclimb cage times, scratching frequency decreased, Zhen Nao Ning group climb the cage wassignificantly reduced, and no significant difference between the Huoxue Xifeng particle dose,high dose group, sibelium group, Zhen Nao Ning group;2. Effects of Huoxue Xifeng method on the content of5-HT, DA, NE in brain tissue of migraine in rats: compared with the model group, Huoxue Xifeng particles, high dose group,brain tissue of rats5-HT, NE, DA content of sibelium were increased (P <0.05), increased5-HT, NE content of Huoxue Xifeng low in rats brain tissue (P <0.05), Zhen Nao Ning groupNE in the brain tissue of rats increased (P <0.05); the effect on the5-HT content, effects ofHuoxue Xifeng granule medium dose group and flunarizine group on its content is obvious,but no significant difference between two groups in effect on; the content of NE, nosignificant differences among the treatment groups on its content; the effect on the DA content,no significant difference between the effects of Huoxue Xifeng middle dose group, high dosegroup and flunarizine group on its contents.Conclusion:1. Using nitroglycerin subcutaneous injection method can be successfullyprepared animal models of migraine.2. The index shows: Huoxue Xifeng method can regulate the changes of behavior andmonoamine neurotransmitters in brain tissues increased.3. Huoxue Xifeng method is effective in the treatment of migraine rats blood stasis dueto wind control card, worthy of further research and development.
