Objective: to observe the Yi Qi Huo Xue Tong Luo Decoction intervene traumatic deep venous thrombosis of rabbit,and influence the content of P-selectin( CD62p)、 PAF and endothelial morphology, and discuss its intervention mechanismMethods: we breed 32 Japan white rabbits a week,we do not find the rabbit death in the same environment. When at the beginning of the experiment, we make the number for 32 white rabbits; according to random number table,we divide them into four groups: Normal control group(group A), the model group(group B), Yi Qi Huo Xue Tong Luo Decoction with low dose group(group C), Yi Qi Huo Xue Tong Luo Decoction with high dose group(group D); A group of normal animal breeding;B group of animals to give such as volume of normal saline(7.0 ml/kg/d);we use intragastric administration to group C and group D one week,before the start of the experiment, group C with7.0ml/kg/d,group D with 28.0ml/kg/d; We extracte experimental rabbit hollow venous blood of each group and stored them in the biochemical test tube, using 3000 r/min of the centrifuge last 6min; then store save the result in the refrigerator- 20 °; according to the instruction book of ELISA kit, ELISA method were used to detect serum P- select element, platelet activating factor levels; We established a traumatic lower limb venous thrombosis model to group B、C、D using the method of He Fei, Zhao Xueling; we used intragastric administration to each group on 2th day after the beginning of making models: Don’t deal with normal control group A,group B with 7.0ml/kg/d NS,group C with 7.0ml/kg/d Yi Qi Huo Xue Tong Luo Decoction, group D with28.0ml/kg/d Yi Qi Huo Xue Tong Luo Decoction for 6days; We extracte experimental rabbit hollow venous blood of each group and stored them in the biochemical test tube, using 3000 r/min of the centrifuge last 6min; then store save the result in the refrigerator- 20 °; according to theinstruction book of ELISA kit, ELISA method were used to detect serum P-select element, platelet activating factor levels again on 8th day; We used color Doppler ultrasound to exceed the formation of LDVT of group B, group C, D group,and random extact one experimental rabbit from group A and group B, C, D,we dissect experiment rabbit and obtain femoral vein blood vessels to make pathological section which HE staining for the femoral vein blood vessels observe the histomorphology. All the data are processed using SPASS17.0 statistical software analysis.Results:1.The gross morphology observation:A week before the beginning of the model of DVT,A group of normal feeding, B group was given saline;Traditional Chinese medicine(TCM) continuous lavage group C, D group; the food—intake,water—intake and activity level of two group rabbits both reduced,but gradually returned to normal on 2th day; 1 experimental rabbit of group B occurrence diarrhoea,died on the second day;1 experimental rabbits of group C in the process of Lavage,the experimental rabbit presented with severe choking vomit lavage and violent resistance, shortness of breath, unresponsive, curled up lazy move, died the next day. There is no rabbit death before the beginning of the model of DVT.The food—intake,water—intake and activity level of group B,C,D rabbits all reduce, they appear Poor mental state, Color dim, vigilance is extremely high questions, but gradually return to normal on 2th day; 1 experimental rabbit of group A occurrence diarrhoea,died on the second day;1 experimental rabbit of group C and 2 experimental rabbits of group D failed to fill the stomach, appeared Severe choking vomit,severe resistance, shortness of breath Slow, curled up lazy move emerging questions when using intragastric administration to them,die on 2th day morning. there is no rabbits death to the end of experiment.0n 8th day,the molding of the lower limbs group B, group C,group D all have different degrees of swell, Yi Qi Huo Xue Tong Luo Decoction with high dose group of swelling degree is the lightest, Yi Qi Huo Xue Tong Luo Decoction swelling degree is slightly high dose group, Swelling in the model group is most severely.2.P- select element, PAF levels:Before the beginning of molding, the level of P- select element,PAF in Yi QiHuo Xue Tong Luo Decoction with low dose group and Yi Qi Huo Xue Tong Luo Decoction no obvious difference was found between normal group and mold group, there is no statistical significance(P > 0.05);After model establishment, Japan rabbit blood P- select element content, PAF content increased obviously, the level of P- select element,PAF of statistically in Model experiment significant differences in the normal group(P < 0.01); Compared with model group, low dose group of experimental rabbit serum P-select(CD62P), platelet activating factor(PAF) content reduced, statistically significant difference(P < 0.01);Compared with the normal group, low dose group of experimental rabbit serum P- choose(CD62P) content is relatively high, no statistically significant difference(P > 0.05), platelet activating factor(PAF) content is relatively high, statistically significant difference(P < 0.05);The P-select content, PAF content of Yi Qi Huo Xue Tong Luo Decoction with high dose group is compared with model group, the content has a statistically significant difference between the significantly lower(P < 0.01); compared with normal group, P- select element content, PAF content moderately elevated, but there was no statistically significant difference(P > 0.05);3. Results of color Doppler ultrasound:In model group 6 experimental rabbit of 7 experimental rabbits have right lower limb thrombosis; Yi Qi Huo Xue Tong Luo Decoction with low dose group with 6 experimental rabbit,there were 1 experimental rabbit right lower limb venous thrombosis; Yi Qi Huo Xue Tong Luo Decoction with high dose group of five experimental rabbit, only 1 experimental rabbit appear right femoral vein thrombosis of lower limbs; Yi Qi Huo Xue Tong Luo Decoction with low dose group, high dose group were lower than the model group the incidence of lower extremity deep vein thrombosis in rabbits.4. Histomorphology observation :Vascular endothelial of Normal group has good continuity, without interruption, defects, blood cell adhesion; Model group endothelial injury is serious, interruption, accompanied by a large number of the defect in the vascular endothelium, fall off, and the fill tube cavity massive thrombosis; the vascular endothelial damage degree of Yi Qi Huo Xue Tong Luo Decoction with low dose group is has part lighter than model group,its vascular intima is rules, part of endothelial interruption and defects;Only a small amount of thrombus adhesionin the lining of blood vessels; Yi Qi Huo Xue Tong Luo Decoction with low dose group has only part of the vascular endothelial injury, loss, It Relatively complete the lining of blood vessels, there is a small amount of blood cell adhesion hemal wall can be seen in the lumen.Conclusion:1. Trauma fixed building method can be traumatic lower extremity deep vein thrombosis model is set up.2.Yi Qi Huo Xue Tong Luo Decoction can effectively Intervene rabbit traumatic lower Limb deep venous thrombosis.3.Yi Qi Huo Xue Tong Luo Decoction can protect vascular endothelial cells, restrain the level of p- select, PAF to inhibit platelet activation, to reduce platelet aggregation,to intervene in the deep vein thrombosis due to trauma.4.
