In this experiment, at first, it was discussed about the extraction process of flavonoids. Through the single factor experiment and orthogonal test, the optimal extraction combination of total flavonoids from Convallaria majalis. was obtained. That is followed:Ethanol concentration is controlled to 70%, Extraction temperature was 80℃、the ratio of materiala and Liquid was1:20(w/v)、 Extraction time was 2h、Extracting for 3 times.In order to get higher purity of total flavone, it was purified by polyamide chromatography, Purification process was researched by static adsorption experiment and dynamic adsorption experiment, It was found that polyamide resin had good adsorbing effect at room temperature and the adsorption solution pH was alkaline; 60% ethanol was best to elution as strippant. It was found that the adsorption rate was 2.0mL/min by the dynamic adsorption experiments, and the concentration of 8mg/mL of flavonoids extracts solution was more appropriate. Elution rate was 2mL/min at room temperature, and40% ethanol was best to elution as strippant. Flavonoids extracts of Convallaria majalis. was dealt with polyamide resin adsorption for 3 times, the content of flavonoid was from 35.40% of crude products rise to 84.52%, the purity has increased 49.12% and was improved by 1.39 times comparing to crude product.In this paper, it was researched that the total flavones of Convallaria majalis. affected to the cardiomotility of toad and rat. In the dectection of cardiotonic effect, it used the simultaneous measurement of heart rate, cardiac output, left ventricular pressure and other indicators. On the cardiotonic effect study on Toad, it show that the total flavones of Convallaria majalis. not only had a role to the normal toad heart failure, it also had the obvious effect on the low calcium induced heart failure of toad. Through the experiment, it was also found that the total flavones of Convallaria majalis. just only have produced the pharmacological effect on the heart failure, made the heart failure gradually returned to normal, but could not produce the high load and oxygen consumption to heart, And with the increasing concentration of reagents, heart rate was slowed down gradually(P<0.05 或 P<0.01), cardiac muscle contraction ability was gradually strengthened. Cardiac output was increased gradually(P<0.05 或 P<0.01), on the research of cardiotonic effect of rat, it was show that total flavones Convallaria majalis. play cardiac effect, at the same time, it did not produce the high load and oxygen consumption to heart, with the increasing concentration of reagents, heart rate was slowed down gradually(P<0.05), Cardiac muscle contraction ability was gradually strengthened.All the results mentioned above showed that:it was sure that the total flavonoids of Convallaria majalis. could improve cardiac dysfunction, and have obviously cardiotonic effects.
