Objective:This experiment by intraperitoneal injection of adriamycin method replication model in chronic heart failure rats and observe the effect of Yangxin Decoction on chronic heart failure rat tumor necrosis factor TNF-α and explore the mechanism of effects of the drug in the prevention and treatment of chronic heart failure, is developed in the treatment of chronic heart failure exhaustion of Chinese Medicine lay the foundation.Methods:90 healthy SD rats, weighing 180-220g, were selected from the experimental animal center of Heilongjiang University Of Chinese Medicine.20 SD rats were taken as blank group, and the other 70 rats as model group,1 were treated by intraperitoneal injection of adriamycin (4mg/kg) times a week for 6 weeks. After 6 weeks, two groups were randomly selected in each of the 10, by color Doppler sonography, LVEF less than or equal to 60% and determination of two groups of rats serum TNF -alevels, make sure the model successfully. The successful SD rats were randomly divided into control group, experimental group and model group. During the treatment, the patients in the control group were given captopril (2.25mg/kg/d) by gavage, the experimental group given Yangxin Decoction (2.34g/kg/d) by gavage, model group and blank control group given the same volume of distilled water, above each day 1,4 consecutive weeks of gavage. In the course of treatment, the general situation of rats was observed, including mental condition,hair, diet, water intake, weight gain and other indicators. After the end of the experiment, determination of cardiac function in rats with left ventricular ejection fraction (LVEF), according to the requirements of blood were taken from the abdominal aorta and ELIS A detection of rat serum TNF -alevels; under the light microscope observation of the rats myocardial pathological morphology changes, and statistical analysis.Results:1.Rats pathological results:blank group, the pathological sections showed normal myocardial cell size, arranged in neat rows, uniform cytoplasm, centerless muscle fiber fracture; model group, the pathological sections showed myocardial cells was significantly larger, arranged in disorder, cytoplasm concentrated dye, myocardial fiber rupture, between the two has significant difference. The experimental group and the control group, the myocardial cells were slightly increased, the cardiac muscle arrangement was neat, the cytoplasm was uniform, the heart muscle fiber breakage, there was no significant difference between the two.2.Each group of big mouse TNF -aresults:compared with the control group compared to model group rats serum TNF alpha levels increased, the difference is statistically significant (P< 0.05); compared with the model group, the experimental group and control group to reduce the serum levels of TNF-a, the difference is statistically significant (P< 0.05); experimental group and control group the difference was not statistically significant (P > 0.05); under the light microscope observation of the rats myocardial pathological morphology changes, and statistical analysis.3.The results of LVEF rats:compared with the blank group, model group EF was significantly lower, the difference was statistically significant (P<0.05). Compared with the model group, experimental group and control group increase the rat left ventricular ejection fraction (LVEF), the difference was statistically significant (P<0.05); the experimental group and the control group are compared, the difference was not statistically significant (P> 0.05).4.Rat heart mass index results:compared with the blank group compared the rats in the model group the heart mass index generally increased, the difference is statistically significant (P< 0.05); compared with the model group, the experimental group and the control group rat heart mass index weight decreased, the difference is statistically significant (P< 0.05); experimental group and control group, the difference was not statistically significant (P> 0.05).Conclusion:1. Yangxin Decoction can reduce the pathological injury of myocardial cells in rats with chronic heart failure.2. Yangxin Decoction can reduce chronic heart failure rats serum TNF-a levels.3.Yangxin Decoction can improve the level of LVEF in rats with chronic heart failure, reduce the cardiac mass index, improve heart function.
