| Objective:To establish a rat model of chronic heart failure by adriamycin in order to observe the effect of Yangxin Decoction on serum NT-proBNP and Ang ?,so to study the mechanism and function of Decoction in Treating Chronic Heart Failure.Methods:60 male rats,weight(200±20)g,10 rats were randomly selected as control group,the remaining 50 rats received intraperitoneal injection of adriamycin 4mg/kg/w,a total of 6 weeks,6 weeks for all except the blank control group without death rats underwent echocardiography,the LVEF<60%to determine the failure model to replicate the success of the standard.All 42 rats were randomly divided into three groups,Yangxin Decoction group,Captopril group and model control group,with each group having a total of 14 rats.A total of four groups of rats for 4 weeks,Yangxin Decoction group given Yangxin Decoction extract 8.1g/kg/d;Captopril was given Captopril 2.25mg/kg/d;the same volume of distilled water were fed to rats in model control group and blank control group.After 4 weeks,the levels of serum NT-proBNP,Ang ? were measured in the abdominal aorta of rats,and the changes of myocardial pathological changes were observed by HE staining.Results:1 The serum NT-proBNP of rats in each group:compared with the blank control group,the serum NT-proBNP level of the model control group,Yangxin Decoction group and captopril group increased,the difference was statistically significant(P<0.01).Compared with the model control group,the serum NT-proBNP level of Yangxin Decoction group and captopril group decreased,the difference was statistically significant(P<0.01).Compared with the captopril group,the serum NT-proBNP level of Yangxin Decoction group was increased,but the difference was not statistically significant(P>0.05).2 The serum Ang ? of rats in each group:Compared with the blank control group,the serum levels of Ang ? were significantly increased in the model control group,Yangxin Decoction group and captopril group(P<0.01).Compared with the model group,the levels of serum Ang ? were decreased in Yangxin Decoction group and captopril group(P<0.01).Compared with captopril group,the levels of serum Ang ? were increased,but the difference was not statistically significant(P>0.05)3 The HE staining of myocardial in each group:the blank group:myocardial fibers arranged neatly,the cell membrane and the nucleus is clear,the stripes are clear and high,no cell edema,no fiber breakage.There was no inflammatory cell infiltration and cell infiltration.In the model group,the myocardial fibers were arranged irregularly,and the fibers were broken,the nuclei were shrun.Conclusion:1 Yangxin Decoction can reduce the level of serum NT-proBNP in rats with chronic heart failure;2 Yangxin Decoction can reduce the level of serum Ang ? in chronic heart failure model rats,inhibit the RAAS system,delay heart failure. |
