Prunus mongolica Maxim.was the endemic species in desert of middle Asia,which played roles of landscape plant,water and soil conservation plant and special species.The plant also plays very important role in stability of the local ecological environment.In addition,being a traditional Chinese medicinal plant and an oil plant as well as an excellent ecological species,P.mongolica also showed very high applying values in its exploitation and utilization.In this article,extraction procedures of seed oil optimized to provide an experimental basis for development and utilization of P.mongolica oil.The main results were as follows:1.P.mongolica seeds oil extracted Soxhlet` method according to national standards(GB/T5009.6-2003).The results showed that crude oil extraction efficiency was ranged from 50.73% to 53.54%.When using ethyl acetate,n-hexane and petroleum ether,the extraction ratios were 53.54%,52.61% and50.73% respectively,which indicated P.mongolica was a high-oil plant.2.P.mongolica seeds oil was extracted with n-hexane and petroleum ether.,the extraction ratio were 92.97% and 86.14% with solvent extraction method;at room temperature,extraction ratio were 95.46% and 86.14% with heating circumfluence method,respectively.It concluded that extraction efficiency with n-hexane was higher than with Petroleum ether.3.Solvent-aqueous enzymatic extraction solved the problem of extract emulsion and fat separation caused by aqueous enzymatic extraction effectively.The extraction ratio would be better under following conditions: denaturing enzyme in boiling water vapor for 5min,stirring for 10 min before enzymolysis,adding organic solvent before enzymolysis,preparing solvent system of petroleum ether and material liquid at the rate of 1:3.4.Analyzing the data of the single factor experiment by using the method of quadrate experiment,the paper obtained the optimum conditions of P.mongolica seed oil extraction:1)Optimum condition of pectinase enzymolysis : enzyme concentration,temperature,time and pH value were 0.5%,25?,3h and 4.5 respectively.2)Optimum condition of cellulase enzymolysis : enzyme concentration,temperature,time and pH value were 1.0%,40?,2h and 5.5 respectively.3)Optimum condition of dispase enzymolysis : enzyme concentration,temperature,time and pH value were 1.25%,40?,3h and 7.5 respectively.As for three Solvent-aqueous enzymatic extraction above,One-time oil extraction rate were 47.87%?48.05% and 52.54% respectively.
