Slightly Acidic Electrolyzed Water(SAEW)is widely applied for its low cost,fast and high efficiency and broad-spectrum antibacterial activity with little remains.But few studies have been worked on its antimicrobial effects.Our study was focused on the antimicrobial mechanism of slightly acidic electrophoresis water.Results were established by comparing the sterilization efficiency between the traditional antibiotics and different material to liquid ratio of SAEW,measuring the changes in membrane permeability,nuclear morphology,cell apoptosis rate and testing the anti-infection effect on Tilapia.The SAEW was made under the parameter of pH 6.0.ORP 890 mV?ACC 28 mg/L.Conclusions were drawn as follows:1)SAEW have strong Sterilization effects on A.hydrophila.Compared to gentamycin and norfloxacin,SAEW has a higher inhibition rate which is up to 96%.Barely a bacterial colony was harvested from the agar gel after treated by SAEW with a material to liquid ratio of 1:20.2)SAEW treatment could induce cell morphology changes in A.hydrophila.With the treating time and material to liquid ratio increasing,the bacterial grown bigger and longer.Hoechst stain on nuclear also showed the same results.3)SAEW can cause cell apoptosis in A.hydrophila.Annexin V-FITC/PI stain showed that membrane permeability increased and apoptotic characteristics were shown after the treatment of the SAEW.The apoptotic rate reached to 40.1%by 1:20 SAEW treatment.DNA ladder also manifest that apoptosis occurred for DNA fragment could be observed at 1200bp.4)In vivo experiment,control group was infected by A.hydrophila while the experiment group treated with SAEW was not.In bacterial infected group,the fish appears black than the normal group,muscle turns pale and eyes bulging out.In the experimental group,the body remains normal,no obvious tubercles discovered and muscle kept fresh.5)Staining slices of fish tissues in control group showed that inflammatory reactions were obtained from the intestine,stomach and liver.Moreover,A.hydrophila infection could break down the basic structure of intestine which meant the function had been lost while stomach and liver does not.This implied that intestine is the easiest to get infected,then comes with stomach and liver is the least to get infected.In the treatment group,no obvious symptoms can be observed.
