objective: Before and after Cauterization Therapeutic Technique(CTT)inpatients with chronic tonsillitis,By observation of the structure of tonsil crypt beta defense-3 change in quantity,To explore the treatment of chronic tonsillitis cauterization of the possible therapeutic theory,And to guide clinical work provides experimental and theoretical basis.Methods: 1.Experimental group:select 15 in a patient with chronic tonsillitis,healthy volunteers 15;Before treatment by CTT,select 15 tonsil tissue samples of 15 cases of chronic tonsillitis patients as the experimental group A(the before CTT groups).After treatment by CTT,select 15 tonsil tissue samples of 15 cases of which have cured of patients withchronic tonsillitis as the experimental group B(the after CTT groups).Healthy volunteers 15,choose the tonsil tissue samples 15 cases in group C the health volunteer group).2.Group A,B,C,making paraffin embedding tissue and pathological specimens,from the morphological observation and immunohistochemical detection beta defense-3.3.Group A,B,C,extract the specimens,total RNA,using reverse transcriptase polymerase chain reaction(RT-PCR)test specimen beta defense-3 in theexpression of transcription.4.Group A,B,C,extract the expression levels of hBD3 proteinin specimens was identified by Western blotting.Results:1.Hematoxylin eosin staining results showed that group A is chronic tonsillitis organization,group B,group C as normal tonsil tissue.2.The semi-quantitive analyses have been shown the hBD3 expression.In the group A,12 specimens are strongly positive,2specimens are positive and 1specimens are weakly positive.In the group B,14 specimens are negative and 1specimens are weakly positive.In the group C,14 specimens are negative and 1specimens are weakly positive.The before CTT groups of hBD3 expression in immunohistochemical quantity obviously higher than the health volunteer group.Choose rank and inspection,statistically significant(P<0.05).The before CTT groups of hBD3 expression in immunohistochemical quantity obviously higher than the after CTT groups.Choose rank and inspection,statistically significant(P<0.05).3.Rt-pcr results showed that A group of about 182 bp fragment was observed,at about 182 bp has A bright,according to A single band and the design of hBD3 fragment length.Group B and group C at about 182 bp has a single gray stripe.The before CTT groups was obviously higher than the health volunteer group.Choose the t test,statistically significant(P<0.05).The before CTT groups was obviously higher than the after CTT groups.Choose the t test,statistically significant(P<0.05).4.Western blotting test results show that molecular weight about 5 kd group A visible deeper stripe,consistent with thedesign of hBD3 protein fragment length.The group B and the group C molecular weight about 5 kd shallow stripe.The before CTT groups was obviously higher than the health volunteer group.Choose the t test,statistically significant(P<0.05).The before CTT groups was obviously higher than the after CTT groups.Choose the t test,statistically significant(P<0.05).Conclusion:1.With the chronic infections in tonsil,which can induce hBD3 from the expression of gene and protein levels rise,to exert its tonsil local immune regulation and antimicrobial function.2.After treatment by CTT to cure chronic tonsillitis,tonsil tissues hBD3 gene and protein expression of decline.3.The CTT therapeutic theory may make the hBD3 decline in the expression of gene and protein levels,and instead of HBD-3 to exert its tonsil local immune regulation and antimicrobial function.
