Objective: High-voltage burns is a special reason of burns,can cause a series of complicated pathophysiologic changes,microcirculation disorder is one of the important pathological changes.Previous animal experiments have found that high voltage causes peripheral tissue microcirculation change not only,and can cause organ of microcirculation disorder apart.Previous animal experiments found that electrical burns after the pia mater of deep fascia,mesenteric,liver,kidney,cortex and pancreas are found in the tissues and organs such as microvascular microvascular diameter reduction,slow blood flow,capillary contraction,microcirculation blood silt up and even the emergence of a micro thrombus,suggests that microvascular blood flow change,platelet aggregation and thrombosis,is the important reason of the change of the microcirculation.Serotonin(5-HT)is an inhibitory neurotransmitter.The tissues,serotonin is a potent vasoconstrictor and smooth muscle contraction stimulants.Catecholamine(CA),including norepinephrine(NA or NE),epinephrine(Ad or E)and dopamine(DA).5-HT and CA is platelets release reaction produced one of the important factors in the process,by activating the surrounding platelets,promote vasoconstriction,fibrous protein to form a variety of ways,such as strengthening the injury of inflammation and immune response,the electrical burns after 5-HT and the change of CA and inflammation,thrombosis,microcirculation dysfunction such as the prevention and control is of great significance.Ulinastatin(UrinaryTrypsinInhibitor,UTI)is a kind of protease inhibitor,can inhibit blood coagulation abnormality hyperthyroidism and secondary fibrinolytic,reduce high coagulation state,as well as reduce platelet release reaction to improve microcirculation.Therefore,we design the experiment,the high voltage serotonin early burn rats,the change of catecholamine expression,and through the ulinastatin(UTI)treatment intervention,research the adjustment mechanism of microcirculation after burn,the study on the feasibility of UTI treatment of high-voltage burns.Methods:1 The experimental animal groups: healthy adult SD rats(provided by the animal experiment center of hebei medical university,certificate number 1211057),180,random number table method is divided into three groups,namely the false high voltage burn group(control group),high voltage burns electrical injury(group),high voltage burns ulinastatin(UTI)in treatment group(treatment group),each group of 60.Each group according to the observation time divide again before the shock immediately after 15 min,electric shock,shock after 1 h,2 h,4 h,8 h six phase group,at 10.2 Before the trial preparation: the number of rats,weighing,left arm,right leg and chest hair removal.Experimental drugs according to the experiment needs match into the required concentration.3 High voltage burn model: good connection test transformer and voltage regulator,press with chloral hydrate the dose of 3 ml/kg intraperitoneal injection with anesthesia experimental rats,after the success of the anesthesia,fixed in experimental rats on stage,in the supine position,the two 1 cm * 1 cm of electrodes were placed in the left upper limb of the rat entrance(current)and the right lower limb(current export)hair removal area.Connection booster,adjust the pressure regulator,the output voltage regulation to 2 kv,connected to the power supply,make the high voltage current is too large rats for 3 seconds.Make fake electrical injury model control group,not closing by electric current,the remaining steps in accordance with electrical injury group.By intraperitoneal injection of electrical injury within 5 min,the treatment group 5 x 104 u/kg UTI,control group and intraperitoneal injection of electrical injury group the same amount of saline solution.Specimen collection and preservation: 4 copies high-voltage burns model rats to the success of open chest exposed heart,look straight into the heart blood 6 ml,of which 3 ml in disposable vacuum blood vessels(in red)upside down gently let stand for 30 min,after several times on centrifuge,in 3000 r/min centrifuge for 10 min,vacuum mining in blood vessels that put in Eppendorf tube supernatant fluid at 70 under the condition of preservation.?5 Index detection: centrifugal separation in the serum of freezer by double antibody sandwich ELISA method,5-HT and the content of CA.6 Experimental data processing: using statistical software SPSS 13.0,line two factors factorial design analysis of variance,multiple comparisons using LSD method t test.With P < 0.05 for the test of significance level.Results:1 The rats serum content of 5-HT changesElectrical injury group 5-HT content in general is higher than the control group(P < 0.05),and 5-HT content over time after injury has significant difference(P < 0.05),showed a trend of increase.Overall treatment group(5-HT content below electrical injury group(P < 0.05),and 5-HT content over time after treatment had significant difference(P < 0.05).2 in the rat serum CA content changesElectrical injury group total CA content was higher than control group(P < 0.05),and the content of CA after an injury over time and there were significant differences(P < 0.05),showed a trend of increase.Treatment group total CA content is lower than the electrical injury group(P < 0.05),and CA content over time after treatment had significant difference(P < 0.05),the treatment group 5 min after injury ~ 8 h each phase are lower than before from head injury when value(P < 0.05)Conclusion:1 The high voltage early burn rats plasma 5-HT,CA expression also increased to different extent,prove the existence of burns after 5-HT,CA,the capillary contraction resulting in microcirculation.2 High voltage burns UTI treatment group 5-HT,CA compared with electrical injury group were decreased,prompt UTI treatment can inhibit the 5-HT,CA secretion,reduce capillary contraction,improve microcirculation,alleviate the body damage.
