Objective:The aim of this study was to explore the mechanism of missed abortion in aspect of angiogenesis by comparing the expression of CLEC14 A and VEGF and ultrasonic Doppler blood flow indices between women with and without missed abortion.Methods: With 30 cases of decidual tissue in missed abortion group(group I)and 50 in control group(group II),using the methods of western blotting and RT-PCR,the difference in expression level of CLEC14 A and VEGF between the two groups were detected.We also observed the uterine blood perfusion by recording RI,PI and PSV/EDV of uterine artery and spiral artery of 5mm around gestation sac.All relevant data were analyzed using SPSS17.0.Results:(1)General information: There was no significant difference(P>0.05)between two groups in age and pregnancy age;RI?PI?S/D of uterine artery on pregnancy sac side,as well as average RI and PI of both side,were significantly higher in group I than in control group(P<0.05).(2)In western blotting,CLEC14 A expressed in the decidual tissue of both group I and group II,which relative CLEC14 A expression were 0.06±0.02 and 0.09±0.05;VEGF expressed in the decidual tissue of both group I and group II,in which relative VEGF expression were 0.49 ± 0.16 and 0.59 ± 0.16.The expression of CLEC14 A and VEGF in group I was lower than group II,the difference was significant(P<0.05).According to RT-PCR results,CLEC14 A and VEGF mRNA expression are also significantly lower in missed abortion group(1.89±0.57 and 1.21±0.50,respectively)than in control group(2.23±0.61 and 1.59±0.66)(P<0.05).(3)Positive correlation was found between VEGF and CLEC14 A in the decidual tissue(r=0.355,P=0.018).VEGF also have negative correlation(r=-0.731,P<0.001)with S/D of uterine artery on pregnancy sac side in group I.Conclusions: VEGF and CLEC14 A expression level,which have positive correlation,decreased significantly in decidual tissue of women with missed abortion compared withcontrol group.It suggested that CLEC14 A might cause missed abortion by regulating VEGF expression to change uterine artery blood flow,especially S/D of uterine artery on advantage side.
