Dryopteris fragrans(L.)schot is a deciduous perennial herb of the Dryopteris genus,which grows mainly in the talc slope of the alpine region,the scree of the forest and lava cracks around volcanoes as small communities.It is mainly distributed in Northeast China,North China,Russia,Japan,Korea,Europe and America.According to the local history records in Wu-da-lian-chi,Dryopteris fragrans' decoction has been used to treat many diseases by Daur people hundreds of years before,and it can reflect the unique advantages of traditional Chinese medicine by its none of side effects.Streptococcus pneumoniae(Streptococcus)is Streptococcus pyogenes which belong to the bacillus outline,lactobacillus eye,Streptococcus families,and widely distributed in the nature.Among it,?-Hemolytic streptococcus possess a strong pathogenicity and can cause a variety of diseases.According to the large number of experimental studies and clinical tests,?-emolytic streptococcus has a close relation with the pathogenesis of rheumatoid arthritis(RA).In recent years,more extensive studies have been done on the chemical composition of Dryopteris fragrans and its treatment of various diseases such as psoriasis and rheumatoid arthritis,but its active ingredients of low polarity have barely been reported.In this paper,indomethacin was treated as the postive control,the ethanol extract of Dryopteris fragrans was extracted by ethyl acetate/water,and the ethyl acetate and water fractions were determined by the antibacterial spectrum of?-Hemolytic streptococcus.The results showed that the minimum inhibitory concentration was 16.625 mg/mL in the ethyl acetate fraction,and 62.500 mg/mL in the water fraction,which presented a significant difference between the ethyl acetate fraction and the indomethacin group.Furthermore,to determine the effective chemical composition of Dryopteris fragrans in treating rheumatoid arthritis caused by?-Hemolytic streptococcus,the effect of ethyl acetate and water fractions on IL-17,TNF,and synovial tissue were studied by microscopy techniques and immune-his-chemistry technology.The results showed that the best therapies for RA were the high-dose ethyl acetate group and the high-dose indomethacin group.Finally,ethyl acetate group was identified as the main active ingredient,.and its chemical compositions were separated.In this paper,the chemical compositions in the purified ethyl acetate fraction of the Dryopteris fragrans were separated and 23 chemicals were achieved by the normal-phase silica gel column,RP(MCI,ODS,RP-18)column,Sephadex LH-20 column chromatography.The structures of 15 chemicals among them were identified by UV,IR,MS,NMR and the determination of various physical and chemical constants,which included 3 sesquiterpenoids,1 coumarins,7 phloroglucinol derivatives,1 sitosterol and 3 lignans.There were 2 new compounds in question that were 2,3-Dihydroxy-1,3,5-cadinatrien-10-Metylformate,8-hydroxyl-4-isopropyl-7-methoxy-6-methyl-2H-benzopyran-2-one.Some chemical compounds which were selinane type sesquiterpene,(+)3-(3',4'-dimethoxy)-4-(3'",4"'-Dimethoxy styryl)cyclohexene,(-)3-(3',4'-dimethoxy)-4-(3'",4'"-dimethoxy styryl)cyclohexene and 4,4'-dihydroxy-3,3'-dimethoxy-7,9':7',9-two epoxy lignin,these 4 compounds were first found in the genus.5 compounds were screened for anti-inflammatory activity.Finally,2',6'-dihydr-oxy-4'-methoxyl-3'-butyl benzene methyl ketone and 2',6'-dihydroxy-4'-methoxyl-3'-m-ethyl benzene acetone were identified that had anti-inflammatory activity.This thesis studied the inhibition effect of Dryopteris fragrans on ?-Hemolytic streptococcus and screened the the monomeric compounds which had anti-inflammatory activity,finally got the active leading compounds,which provided new medicinal resource for the treatment of diseases by ?-molytic streptococcusin the future.
