Entrepreneurship is the driving force of social and economic development, and the regional economy usually develops rapidly during the period of entrepreneurial activity. We found that in the rapid development of entrepreneurial activities, entrepreneurs in the choice of business location often gathered in the same area, which is the phenomenon of entrepreneurs get together,an important reason for entrepreneurship is knowledge overflow. This paper is based on economic geography to discuss entrepreneurship, systematically study the regional differences of entrepreneurial activity from the perspective of knowledge spillovers. After the introduction of the entrepreneurial system approach, this paper constructs a three-stage structural model for the regional entrepreneurial system. The model is based on the theory of knowledge spillovers, the regionalization of knowledge spillovers, and the regionalization of human capital.A source of opportunity to regard human capital as the main source of entrepreneurial absorptive capacity. And based on the data of the Pearl River Delta to test, using the path analysis method to explore the relationship between knowledge, entrepreneurship and geography. The empirical results show that knowledge spillovers are there, but knowledge spillovers do not have any weight in the whole entrepreneurial activity.Even for high-tech industries, the impact of new knowledge on entrepreneurship is not significant, due to the existence of knowledge filters.Human capital plays a key role in entrepreneurship, but its role is mainly to increase the ability of entrepreneurs to absorb rather than create knowledge-based entrepreneurial opportunities.Industrialization, university and social diversity are regarded as important predictors of regional human capital stock. The overall impact of university on entrepreneurial activity is negative.Industrial specialization and social diversity are positive for entrepreneurship. Thus, the local public policy, especially at the city level,is to build an entrepreneurial economy, primarily to create a regional environment that attracts human capital, emphasizing that non-discrimination,fairness and democracy encourage social diversity while public policy should focus on dredge the channels of knowledge spillovers, break through the regional “knowledge filters”,but also encourage the development of high-tech industries, thus bringing more entrepreneurial and innovative regional economy.
