Rifamycins are a class of antibiotics which used for treating chronic infectious diseases,tuberculosis.Tuberculosis has been plaguing human's health since its appearance.There are many people with AIDS suffer from tuberculosis in Asia and Africa.Therefore,the study of such drugs for the treatment of tuberculosis has a positive effect.The chemiluminescence method as a rapid analysis of detection methods,not only has high sensitivity,but also easy to operate.As a research hotspot,the flow injection chemiluminescence?FI-CL?is widely applied in medicine,health,environment and food testing in recent years.This thesis is divided into two parts.The first part summarizes the background,structure and research significance of rifamycins with the methods commonly used to determine the content of rifamycin antibiotics.It briefly introduces the production of flow injection analysis technology.Besides,I have made a summary to the analysis methods commonly used with flow injection.Finally I have introduced the basic principles of chemiluminescence and the application of flow injection chemilumi-nescence in various types of organic or inorganic quantitative analysis.The second part is the research report.I have set up several new methods combined with flow injection technology through the basis of the optimal experiment condition to detect the content of rifapentine,rifabutin,rifaximin and rifampicin by using the Luminol chemiluminescence system and KMnO4-formaldehyde system.I have applied these new methods to analysis drug content in real table capsules.The details are as follows:1.A novel method combined with flow injection analysis technology for the determination of rifapentine was proposed based on the phenomenon that the chemiluminescence intensity of the Luminol-K3Fe?CN?6 system could be enhanced by rifapentine in alkaline medium.Under the optimal experimental conditions,the linear range of rifapentine's detection concentration was 8.0×10-30.88 mg/L and the detection limit?3??was 1.2×10-3 mg/L,the relative standard deviation?RSD?was 1.9%?n=11?for rifapentine at 5.0×10-2 mg/L.The recovery rate is in the range of 98.6%103.7%.The method has been applied to the determination of rifapentine in capsules.2.A new method combined with flow injection analysis technology for the determination of rifabutin rapidly was proposed because the chemiluminescence intensity of the Luminol-KMnO4 system could be enhanced with the adjunction of rifabutin in alkaline medium.Under the optimal experimental conditions,the linear range of rifabutin's detection concentration was 8.5×10-35.9 mg/L and the detection limit?3??was 2.3×10-3 mg/L,the relative standard deviation?RSD?was 1.6%?n=11?for rifabutin at 5.0×10-2 mg/L.The recovery rate is 99.8%102%.The method has been applied to the determination of rifabutin in capsules with satisfactory results.3.The study found that rifaximin could enhance the luminol-hydrogen peroxide chemiluminescence intensity in alkaline conditions and the chemiluminescence intensity within a certain range with the concentration of rifaximin solution was linearly correlated.Based on this phenomenon combined combined flow injection analysis,the experiment explored a rapid and efficient method for the determination of rifaximin.After the optimization of experimental conditions,the linear range of rifaximin's detection concentration was 2.4×10-37.86×10-22 mg/L and 0.233.93 mg/L,the detection limit?3??was 8.0×10-4 mg/L,the relative standard deviation?RSD?was 0.56%?n=11?for rifaximin at 5.0×10-2 mg/L.The recovery rate between 96.7%102%.4.It was found that potassium permanganate can oxidize formaldehyde to produce a weak chemiluminescence signal and adding a certain concentration of RFP solution can enhance the signal.A new chemiluminescence method for the determination of rifampicin was explored in combination with flow injection analysis.After optimization of the experimental conditions,the linear range of rifampicin was 8.2×10-28.23 mg/L and the detection limit?3??was 5.1×10-22 mg/L.The relative standard deviation?RSD?was 2.2%?n=11?for rifampin at 0.5 mg/L.The method has been used to determine the content of rifampicin capsules,the recovery rate was between 98.7%102.7%.
