Extraction And Preliminary Study Of Ganodernza Lucidum Melanin

Due to its toxic side effects,melanin synthesis has been restricted or even banned in some countries Natural melanin is extracted from plants,animals and microorganisms bodies and metabolites,currently mainly from plants.However,due to the slow growth of plants,subject to greater environmental impact and other factors,resulting in insufficient production capacity of natural melanin in short supply Ganoderma lucidum with fast growth,less affected and has long been the realization of factory cultivation,suitable for the production of natural melanin.In this paper,ultrasound-assisted extraction of Ganoderma lucidum melanin,and its structure and biological activity,further applied to the composite beverage,has been studied.The results are as followsSingle-factor test and response surface method were used to optimize the ultrasonic-assisted extraction of natural melanin from Ganoderma lucidum.The optimal extraction process aare as follows material-liquid ratio 1:39.7 g·mL-1,ultrasonic power 160 W,and ultrasound time 49.8 min.Ultrasound power has the greatest impact on the extraction efficiency.Using this process for verification,the absorbance was 0.782,which was 31.42%higher than that before optimization.This shows that this method can effectively improve production effficiency and save costsThe ganoderma lucidum melanin was further purified by Sephadex gel chromatography to give a single fraction G2.The maximum absorption wavelength of this melanin is 232 mn,which is easily soluble in alkaline solution,slightly soluble in distilled water,insoluble in hydrochloric acid,anhydrous ethanol,75%ethanol,n-butanol,isopropanol,and chloroform.Ganoderma melanin has absorption peaks at wave numbers of 3610 cm-1,2952 cm-1,1720 cm-1,1221.82 cm-1,and 1643 cm-1,presumably containing-OH,-NH,C=O,-COOH,and C-H,O-H,indole ring and other structuresBy studying the stability and oxidation resistance of Ganoderma lucidum melanin,it was found to have good thermal stability,light stability,and alkaline stability.It is not suitable for long-term ultraviolet radiation and coexistence with heavy metal ions.Ganoderma lucidum melanin has a strong hydroxyl radical scavenging ability,superoxide anion scavenging ability and metal ion chelation capacity,is expected as a natural antioxidantMangosteen,Ganoderma lucidum melanin,honey and baking soda as raw materials for the composite drinks.Optimized,the best ratio of mangosteen 2.5%,honey 2.94%,Ganoderma melanin 2.83%and baking soda 0.01%,with a sensory score of 90.The beverage is refreshing and delicious with pleasant scent and is suitable for further promotion.
