Viburnum sargenti Koehne,as one of the four famous flowers and sightseeing-worth trees in northeast China,is deeply loved by the city landscaping because of unique shaped and fascinating landscape.As an important medicinal plant,it has a variety of pharmacological activities,and is the raw material of many Chinese medicine products.Therefore,it has high application value in both landscaping and medicinal production.This paper research the propagation of Viburnum Sargenti seeds breeding,Cuttage propagation and tissue culture,The effects of different temperature stratification and different concentration of Gibberellin on seed germination were studied;The effects of different hormone concentrations on the cuttage of hard branches and twigs were investigated;The effects of different sterilizing treatment,different hormone types and concentration on the adventitious bud,leaf callus and adventitious rooting induction of stem segments were investigated;The effects of different kinds and concentrations of antioxidants on Browning were discussed in view of the browning phenomenon in the process of culture.The main results were as follows:1.Using the seeds collected in the year as material,Draining the sand after 5%baking soda.Alternate treatment of high temperature and low temperature,The germination rate is over 52%;2.Using the Viburnum softwood and hardwood as material,The 10 s shoots were treated with 100 mg L-1 IBA,and the survival rate was 61.53%.The 2 h survival rate was 100 mg·L-1 IBA,and the highest survival rate was 53.28%;3.The outdoor collection young branches of Viburnum sargenti were taken as initial material,The optimal aseptic processing condition was sterilized with 70%ethanol and 3%sodium hypochlorite for 20 s and 10 min,respectively.Then used sterile water washing three times and dried the surface of the twigs with sterile filter paper.The twigs were inoculated on WPM medium and the pollution rate was 0;4.Through single factor and orthogonal experiment,established the Viburnum tissue culture regeneration system:The medium for adventitious bud differentiation of stem was WPM + 2 mg-L-1 6-BA + 0.2 mg·L-1 IBA,the differentiation rate was 93.66%;The medium for adventitious bud proliferate was WPM + 2 mg·L-1 6-BA + 0.25 mg·L-1 IBA,the proliferation multiples reaches 7.39,the subculture period is 35 d;The medium for induce callus was MS +2 mg·L-1 6-BA+ 0.1 mg·L-1 IBA+ 2 mg·L-1 2.4-D.the inducement rate was 93.33%?Adding 50 mg·L-1 VC,50 mg-L-1 AC?the browning of the leaves can be reduced;The rooting medium was WPM + 1.5 mg-L-1 IBA + 1 g·L-1 AC,Adventitious buds began to root for 10 days of inoculation,the rooting rate was 94.62%,the number of shoots and the length of buds on average was 8.12 and 4.24 cm;Transplant the seedlings after 2 days,the survival rate reached 100%.
