With the continuous deepening of economic,green,environmental protection and ecology in landscape industry,colorful leaf plants show more and more broad market application prospects,especially colorful leaf tree species with rich color leaves.Acer rubrum L.October Glory is a large deciduous tree of the east coast of the United States of America.Its autumn leaves are Red and bright,with fluorescence.The tree species can not only adapt to a variety of soils,but also have ecological characteristics such as moisture resistance,cold tolerance,drought resistance and absorption of chlorine gas.However,the number of tree species in the country is still scarce,especially the large trees,which has caused a major obstacle to its landscaping.Therefore,in order to scale production of Acer rubrum L.October Glory seedlings,cutting rapid propagation and transformation of rapid propagation technology has become the most effective way.However,due to the high content of tannin and phenolic compounds in the modified red maple plant,the root formation was inhibited,so the root was influenced by the cutting roots,and not only the same difficulty in the tissue culture,but also the key to the formation of the tissue culture.The key to the development of the garden was to break through the technical difficulties of cutting the fast propagation and the rapid propagation of tissue culture.In this way,that red maple tree with the long lobe and the stability of leaf discoloration is the experimental material in North America,and the propagation of the red maple in North America discussed in Acer rubrum L.October Glory.The main research contents and the results have the following several aspects:1.It was observe that that most stable colour change were observed by observe the changes of the red maple in North America,leaf color parameter a*,and the content of anthocyanin in the leaves.It found that discoloration period of October was 1 month earlier than that of the other two types of Red maple.At the same time,there are also differences between the three kinds of Red maple a*values.In the two stages of the early stage of color change,a*is significantly higher than the wine red and autumn flame,and the leaf color also starts to turn red.October to the first?stage,shining a*value is positive,and autumn flame and red wine is still negative.Therefore,Red maple color brilliance"October"in leaf and leaf color parameters on parameter a*value were greater than and autumn flame red wine,and has high ornamental value.In the whole process,the content of total anthocyanin in red maple leaves in North America is changed as follows:October brilliant>wine red>autumn flame.It found that that octobe light has better adaptability.So,we finally according to the laid color performance and stability of leaf flower pigment contents choice brilliance"October"varieties as experimental material.2.North American maple cuttings rapid propagation research:the test through the comparisons of different substrates,growth regulator and concentration,and combined with the cuttings rooting mechanism of NAA and TDZ treatment,study the north American maple brilliance"October"cutting rapid propagation technology.It was found that the root was best in T4?humus soil:quartz sand=3:1:1?.Through the analysis of 5 kinds of rooting agent on the effects of the cuttings rooting found NAA could effectively promote the induction and formation of adventitious roots,the root growth also has the very good effect,and although the TDZ does not stimulate the adventitious root formation of effect,but it can effectively increase the callus formation.The concentration of suitable hormone is different from its cutting effect,which also has a great influence on the rooting rate of north American red maple.NAA had the highest concentration of 15mg·L-1,significantly higher than IBA and IAA.It found that cuttage was the best in winter and spring.Through the analysis of the cuttings of TDZ and NAA treatment,the change of the internal nutrients and related enzymes was changed.And we found that NAA treatment significantly reduced,soluble sugar content in branches after they showed a trend of steady,but TDZ soluble sugar content in the branches of the processing have been reduced;The changes of soluble protein content in the base of NAA and TDZ were both decreased until 16d.The activity of POD activity in the basal POD of the basal POD of TDZ and NAA increased with time,and the activity of POD was slightly higher than that under the treatment of hormone NAA.Hormone TDZ and hormone NAA treated the activity of CAT enzyme in the spike after decreasing,and the activity of CAT was slightly higher than that of TDZ.In this experiment,NAA and TDZ treatment have less effect on the PPO,PPO enzyme activity at the beginning of the cuttings activity decreased significantly.That can make the north American maple cuttings branches respiration,reduces and change regularity:the trend of IAAO activity weakened after first increases,and IAAO enzyme activity peaks appear in the callus formation.3.Study on the rapid propagation of Red Maple leaves:this experiment was conducted to effectively disinfect the external implant by using 4%sodium hypochlorite disinfection for 7min+0.1%Hg treatment of 8min+75%alcohol treatment of 50%alcohol treatment.Leaves and bark explant in early physical disinfection should be treated differently,large water flush the blade speed should not be used to in order to avoid damage the blade,but stem branches explant is different,must through the flow velocity of water flushing to achieve disinfection effect.In the simulated medium environment,1.5mg·L-1 citric acid solution inhibited the Browning effect best.This study found that appeared in the twigs stem section,yellow-green globular?type callus,transgenerational when multiply faster and easy formation of embryonic callus.It was found that the optimal callus culture medium was MS+0.8mg·L-1TDZ+1.0mg·L-16-BA+0.5mg·L-1NAA culture medium,and the recovery rate was 82%.Embryonic callus induction process,optimizing breeding rate faster and grow well?as material of embryonic callus induction,callus cultured in MS+0.6mg·L-1TDZ+0.5mg·L-16-BA+2.0mg·L-1IAA+35g·L-1 to get embryonic callus.The induction rate was the highest in MS+1.0mg·L-1TDZ+3.0mg·L-16-BA+0.2mg·L-1NAA+1.2mg·L-1IAA,up to52%.Theinductionratewasthehighestin MS+0.6mg·L-1TDZ+1.0mg·L-16-BA+3mg·L-1NAA+35g·L-1+7.5g·L-1 agar,and the induction rate was up to 76%.
