Ulcerative colitis is a chronic disease of unknown etiology,Chronic disease characteristics to colonic ulcerative inflame mation[1],Chinese medicine treatment prospects,Clinical experience of matrine of Sophorae mixture enema effect[2]?Our previous studies have shown:General flavone is one of the effective components of matrine of Sophorae mixture in the treatment of ulcerative colitis,further study found that total flavone has bad water solubility,that will directly affect the drug absorption in vivo and the effect of the play.Therefore,this study intends to carry out a comprehensive physical and chemical properties,general flavone analysis method,component separation,quality standard etc.In order to clear the components and characteristics of total flavonoids of Sophora flavescens;the establishment of quality standard of general flavone;to investigate the phospholipid complex technology to improve the total flavone of bad solubility possibility;to lay a foundation for its clinical application.First of all,the preformulation study of total flavonoids of Sophora flavescens,with Sophoraflavanone G as control sample,Using Mg-HCl colorimetry to determine the content of total flavones in Sophora flavescens Ait.Result,the regression equation of Sophoraflavanone G is Y= 0.0253X + 0.0004,R2 = 0.9994;the Linear range is1 0.04?30.1 2?g/ml,The average content of total flavonoids that was calculated by external standard two-point method is 71.8±0.68%(n = 5).To further investigate the solubility of total flavone in different solvents and the apparent oil-water partition coefficient.The results showed that the total flavonoids of Sophora flavescens is poor solubility in water,that belongs to soluble substances according to 2010 version of the two Chinese Pharmacopoeia appendix for drug solubility evaluation standard.The apparent partition coefficient of general flavone is low,that will affect the in vivo dissolution,absorption and curative effect.Second,this paper analyzed the component separation and identification of flavonoids of Sophora flavescens.By silica gel column chromatography,preparative TLC,recrystallization etc,we get 8 compounds.Combined with NMR,UV,IR,MS and technology for structure analysis,and carries on the comparison with the literature data,and ultimately determine the structure of five compounds,they are Sophora flavanone G,iso-anhydroicaritin,isoxanthohumol,formononetin,trifolirhizin.The third.In this paper,the high performance liquid chromatography(HPLC)method to establish the total flavonoids from Sophora flavescens fingerprint,and the evaluation of the similarity of chromatographic fingerprint.On the basis of this,by compared with known compounds,the fingerprint of total flavonoids from Sophora flavescens in five main chromatographic peaks were identified,and of these 5 kinds of flavonoids content determination method was studied,based on the standard curve.The linear regression equation and correlation coefficient and the concentration range of 5 componen are respectively:Trifolirhizin Y =1 × 107X+5099.2,R2=0.9999,linear range 0.0501?0.4008 mg/ml;formononetin Y=4 x 1 07X-5623 81,R2 = 0.9999,linear range 0.0502?0.40 1 6mg/ml;isoxanthohumol Y=4×107X-49812,R2=0.9999,linear range 0.0502?0.4016mg/ml;iso-anhydroicaritin Y=4×107X-671042,R2=0.9999,linear range 0.0504?0.4032mg/ml;Sophora flavanone G Y=3×107X+ 378501,R2=0.9997,linear range 0.0502?0.4016mg/m.Using external standard two-point method is used to calculate the five kinds of compounds in the average content of total flavonoids in Sophora flavescens were trifolirhizin 3.66±0.04%(n = 3),formononetin 2.22±0.01%(n=3),isoxanthohumol 7.60±0.02%(n?3),iso-anhydroicar itin 3.28±0.02%(n = 3),Sophoraflavanone G 6.59±0.03%(n = 3).The method has good precision and reproducibility,providing reference for the quality control of total flavonoids from Sophora flavescens.The fourth.In this paper,the soybean phospholipids of general flavone phospholipid complex was prepared,the recombination rate of the total flavonoids from Sophora flavescens and phospholipid as the evaluation index,on the basis of single factor experiment using orthogonal experimental design,the preparation condition of general flavone phospholipid complex optimized.The best preparation process:The reactant concentration of 2mg/ml,drug and phosphor lipid molar ratio was 1:1.5(w/w),the reaction time is 2h,the reaction temperature is 40 ?.Further research of general flavone phospholipid complex physicochemical properties,general flavone and flavonoids phospholipid complex at different pH value Solubility of phosphate buffer solution and apparent oil-water partition coefficient are studied.The results show that:Total flavonoids from Sophora flavescens and phospholipid compound and solubility range by 0.08?8.4 5mg/ml increased for 0.092?9.325mg/ml,apparent oil-water partition coefficients ranging from 6.54?16.32 increased 11.32?32.87.The total flavonoids in Sophora flavescens,total flavone phospholipid physical mixture and total flavone phospholipid complexes were characterized by DSC,FTIR,UV,and other means.The results show that the total flavonoids of Sophora flavescens and soybean phospholipid was complex,the formation of a new phase,did not form a new compound.According to Chinese Pharmacopoeia 2010 Edition 2 appendix XC provisions of in vitro drug release determination method of comparative study on the total flavonoids from Sophora flavescens and total flavonoids phospholipid complex in vitro release.Examines the 360 release,results of total flavone in Sophora flavescens made phospholipid complex its release has increased to a certain extent,released by 77.8%to 86.4%in 360min.
