Experimental Investigation On The Effect Of Chai Shao Ru Pi Pellet On The Expressions Of Bmi-1 And P63 Proteins In Breast Precancerous Lesion Of Rabbits

Objective:To observe the effect of Chai Shao Ru Pi Pellet on the expressions of Bmi-1 and P63 proteins in rabbits breast precancerous lesion model,and to preliminarily explore its mechanism for breast precancerous lesion.Methods: Out of 38 New Zealand female rabbits,a total of 10 were randomly selected as blank controls and the rest were as modeling preparation group whose modeling was established using Dimethylbenz(a)anthracene in combination with estrogen and progesterone in sequence 5 days method.After two months of modeling,one rabbit was randomly sacrificed in each group to verify the success of modeling.Afterwards,the modeling preparation group was randomly divided into three subgroups:model group,Chai Shao Ru Pi Pellet group,and tamoxifen group,given intragastric administration of normal saline for 20 ml daily,Chai Shao Ru Pi Pellet for 0.94g/kg daily and tamoxifen for0.47g/kg daily,respectively,for one month intervention.After sacrificed,the morphological features of second pair of breast tissue from rabbits was observed by HE staining,and its expressions of Bmi-1 and P63 proteins was measured by immunohistochemical SP method.The data of the study was analyzed by SPSS17.0 software.Results:(1)The drug intervention groups had better results in incidence rate of breast precancerous lesion,the positive rates and average gray values of positive targets of Bmi-1and P63 than the model group(P?0.05).(2)No significant differences were found in incidence rate of breast precancerous lesion,the positive rates and average gray values of positive targets of Bmi-1 in Chai Shao Ru Pi Pellet group,and tamoxifen group(P>0.05),but tamoxifen group had better results in the positive rates and average gray values of positive targets of P63(P<0.05).Conclusions: The result suggest Dimethylbenz(a)anthracene in combination withestrogen and progesterone in sequence 5 days method can establish rabbit models of breast precancerous lesion.Chai Shao Ru Pi Pellet can partly block and reverse breast precancerous lesion,inhibit the expression Bmi-1 protein,and promote P63 protein expression.It could inhibition of proliferation and induction of apoptosis.So it plays an important role in the prevention and treatment of breast precancerous lesion.
