G.lucidum lucidum polysaccharide has lots of biological activities,it can regulate the immune system,improve the host immunity,anti-tumor,ability of blood oxygen,accelerate blood circulation,eliminate free radicals,antioxidant,ant-aging.Previous reports indicated that Fuc-enriched G.lucidum lucidum polysaccharide fraction can induced antibodies recognizing Globo H-series structures and stimulates the B1 B-Cell activation to activate the immune response of the host to achieve the goal of early prevention and inhibiting of the tumor cell growth.We initated to change the medium composition during the fermental cultivation of the Ganoderma lucidum to enhance the fucose composition of Ganoderma lucidum in order to improve the ability of anti-tumor and biological activity.In this study,We based our study according to the fucose metabolic pathways,denovo pathway and salvage pathway,We tried to add fucose,Mannose or fucoidin into the medium to study whether it could increase the content of fucose in Ganoderma lucidum polysaccharide and investigated the best additive and additive amount which has the highest polysaccharide contents of fucose.The results showed that,adding fucose or Mannose into the medium could not increase the content of fucose in Ganoderma lucidum polysaccharide,but adding fucoidin could increase the content of fucose obviously.In order to further study its physical and chemical properties,we used DEAE-cellulose 52 chromatography and Sephacryl S-300 High Resolution to isolate the cure polysaccharides.And the content of fucose in Ganoderma lucidum polysaccharide when adding the fucoidin at amount of 0.8 g/L,1.6 g/L,2.4 g/L,4.8 g/L,6.4 g/L into the medium.The major functional fraction of Ganoderma lucidum polysaccharide were studied through multi-angle laser light scattering(MALLS)and IC.The results showed that,when adding 4.8 g/L fucoidin into the medium is the best additive amount.The content of fucose in Ganoderma lucidum polysaccharide increase from 1%to 17%.The main component of normal meidum Ganoderma lucidum polysaccharide is 94.6%,the molecular weight is 1.262×104Da.The main component of Fuc-enriched Ganoderma lucidum polysaccharide is 82.4%,the molecular weight is 1.233×10~4 Da?In the study of biological activity,We conducted antitumor test in vivo,antioxidant activity test in vitro and cell toxicity experiment respectively to evaluate biological activity between the Fuc-enriched Ganoderma lucidum polysaccharide and the normal control group.The results showed that,Fuc-enriched Ganoderma lucidum polysaccharide has higher antitumor activity than the normal control group.The tumor inhibition efficiency increased from 37%to 58%,by 21%at the concentration of 300 mg/kg.The Fuc-enriched Ganoderma lucidum polysaccharide has better antitumor activity than the normal control group.In the study of antioxidant activity,We detected DPPH radical scavenging ability,hydroxyl radical scavenging ability,the ultra oxygen anion removal ability and reducing power.The results showed that,Fuc-enriched Ganoderma lucidum polysaccharide has higher antioxidant activity than the normal control group.The Fuc-enriched Ganoderma lucidum polysaccharide has better biological activity than the normal control group.
