Acne is a chronic inflammatory skin disease that occurs in hair follicles and sebaceous glands,often occurring on the face,chest,back,and other areas of the body that are rich in fat.When it occurs,it is usually small papules and purulent sputum,or blackheads.It can squeeze out powdery slag,and it is accompanied by nodules,cysts,scars,pigmentation,etc.,accompanied by sequelae such as hypertrophic scars.The GBD(The global burden of disease)study found that the incidence of acne among young people aged 12-25 was as high as 85%.Acne can seriously affect a person's appearance,leading to loss of self-esteem and even mental illness.At present,acne is mainly treated by topical treatment or holistic treatment using antibiotics such as clindamycin,isotretinoin,and benzoyl peroxide and sebacic acid.These drugs have certain therapeutic effects,but also cause many adverse reactions.In order to solve the problems of antibiotics and hormonal drug resistance and adverse reactions,it is urgent to find new and natural drugs that can replace antibiotics to control acne diseases.Wetland plants may play a unique role in resisting pathogenic microorganisms and treating acne due to the particularity of their living environment.Therefore,this research selected 14 species of wetland plants as subject,they were Trapa bispinosa,Euryale ferox,Ceratophyllum demersum,Taraxacum mongolicum,Alternanthera Philoxeroides,Solanum nigrum,Lysimachia christina,Rubia cordifolia,Ranunculus sieboldii,Spiranthes sinensis,Polygonum hydropiper,Polygonum hydropiper,Glechoma longituba and Pisitia stratiotes.The active ingredients were extracted by ethanol extraction to determine their antibacterial activity against Propionibacterium acnes and Staphylococcus epidermidis,and the plant extracts with strong antibacterial activity were screened.Further extracting and separating with different solvents,selecting the extract with the strongest bacteriostatic ability,analyzing the main active substances by LC-MS,and then further studying the antibacterial activity of the active substances against acne-producing bacteria,and studying different extracts on rats The in vivo therapeutic effect of the acne model.To provide a reference for the development of natural,safe and effective alternative antibiotics for the treatment of acne drugs.The results were as follows.1.The ethanol extracts of 7 plants inhibited different bacteria.The extaracts of Trapa bispinosa,Euryale ferox,Alternanthera Philoxeroides,Lysimachia christinae,Spiranthes sinensis,Stellaria media and Lysimachia christinae had a certain inhibitory effect on Propionibacterium acnes and Staphylococcus epidermidis.The ethanol extracts of 5 plants inhibited different bacteria.The extaracts of Taraxacum mongolicum,Rubia cordifolia,Ranunculus sieboldii,Glechoma longituba and Pisitia stratiotes showed the difference between pathogenic bacteria,and only had a certain degree of antibacterial activity against Staphylococcus epidermidis.Among them,2 kinds of ethanol extracts of Ceratophyllum demersum and Solanum nigrum have no antibacterial activity against both bacteria.The concentration of 100 mg/mL stalk ethanol extract and citrus alkaloid extract had a good inhibitory effect on Propionibacterium acnes and Staphylococcus epidermidis,and their inhibition zone diameters for P.acnes were(19.75±0.50).)mm,(20.25 ± 0.96)mm,and the inhibition zone diameters of Staphylococcus epidermidis were(18.25 ± 1.71)mm and(14.50 ±1.00)mm,respectively.2.The 4 extracts of wetland plant Trapa bispinosa have antibacterial activity against Propionibacterium acnes and Staphylococcus epidermidis.The antibacterial activity of ethyl acetate extract against Propionibacterium acnes and Staphylococcus epidermidis is significantly higher than that of petroleum ether extract,n-butanol extract,water extract.The minimum inhibitory concentrations of the 4 extracts against Propionibacterium acnes were 6.25 mg/mL,3.125 mg/mL,6.25 mg/mL,12.5 mg/mL,and the minimum bactericidal concentrations were 12.5 mg/mL and 6.25 mg/mL,respectively,12.5 mg/mL,25 mg/mL.The minimum inhibitory concentrations of the 4 extracts against Staphylococcus epidermidis were 6.25 mg/mL,3.125 mg/mL,6.25 mg/mL,6.25 mg/mL,and the minimum bactericidal concentrations were 12.5 mg/mL and 3.125mg/mL,respectively,12.5 mg/mL,12.5 mg/mL.3.Analysis of petroleum ether extract,ethyl acetate extract,n-butanol extract and water extract by LC-MS,a total of six main components were determined,which were gallic acid,caffeic acid,4-Hydroxycinnamic acid,Isoquercitrin,Cynaroside and Quercetin-3-rhamnoside.The 4 extracts contained gallic acid,and the number of peaks in the mass spectrogram of the ethyl acetate extract was significantly more than the other three extracts,and the gallic acid content was the highest,reaching 2.5358%.However,the 6 polyphenolic substances have no antibacterial activity against Propionibacterium acnes and Staphylococcus epidermidis.4.After treatment for two acne models for 14 days,IL-6,IL-8 and TNF-? were detected in the serum of the ethanol extract of the ethanol extract of the group.The results showed that the ethanol extract can inhibit the pathogen of acne,and its ethyl acetate extract has the best antibacterial effect on acne pathogenic bacteria,but its polyphenols are not the main component of antibacterial activity,acetic acid.Ethyl acetate extract as a topical treatment for acne medications can significantly reduce inflammatory acne conditions.
