Objective:To observe the effects of Jiaji electroacupuncture combined with nerve mobilization on nerve function,CD34,VEGF expression level and blood nerve barrier after sciatic nerve injury in rats,and to explore the related effects of Jiaji electroacupuncture combined with nerve mobilization on peripheral nerve injury Mechanism to provide theoretical basis for clinicalMethods:Randomly divide 135 SD rats into normal control group,model control group,Jiaji electroacupuncture group,nerve mobilization group,and spinal nerve mobilization group(referred to as combined treatment group),27 rats in each group;,2 weeks,4 weeks of treatment time is divided into three subgroups,9 in each subgroup.The use of hemostatic forceps to cause Sunderland(Sunderland)third degree injury model.The model control group is placed in a cage after operation.Like the normal control group,it is taken out every day to appease without any treatment.The Jiaji points of the spinal cord segment were treated with Jiaji electroacupuncture;the combined treatment group was first treated with Jiaji electroacupuncture followed by neuromobilization.The treatment method was the same as above.The above treatments were treated six days a week with one day off.According to each time point,after the sciatic nerve function index(SFI)was used to evaluate the function of the sciatic nerve,the rats in the corresponding subgroup were sacrificed.Observe the count of myelinated axons;take the corresponding spinal cord segments of the sciatic nerve and determine the expression of VEGF by immunohistochemistry;take 3 animals from each subgroup before taking the material and take the sciatic nerve tissue after injection with Evans blue and observe the blood under the microscope Changes in the degree of nerve leakage.Result:1.Sciatic nerve function indexAt the 1-week time point,the model control group,the three treatment groups had lower scores than the normal control group,and the than the normal group,and the combined treatment group was higher than the single treatment group(P<0.05),of which the sciatic nerve was at the 4 week time point The index is the highest.2.sciatic nerve myeloid axon countAt three time points of 1,2,and 4 weeks,the myeloid axon counts of the model control group and the three treatment groups were lower than that of the normal group at each time group reached the peak at the 4-week time point3.CD34 expressionAt three time points of 1,2,and 4 weeks,the CD34 expression of the model control group and the three treatment groups at each time point was higher than that of the normal control group(P<0.05);at the 1 week time point,the expression of Jiaji electroacupuncture group was higher than The model group,neuromobilization group and combination treatment group(P<0.05);at the two and four week time points,the expression of the three treatment groups was higher than the model group,and the expression level of the combination treatment group was higher than the other groups(P<0.05),the expression of CD34 reached the peak at 1 and 2 weeks,and began to decrease at 4 weeks4.VEGF expressionSciatic nerve:at three time points of 1,2,and 4 weeks,the VEGF expression of the model control group and the three treatment groups at each time point was higher than that of the normal control group;at 1 week time point,the expression of Jiaji electroacupuncture group was higher than that of the model group,Nerve mobilization group and combined treatment group;at the two and four-week time points,the expression of the three treatment groups was higher than that of the model group,and the expression level of the combined treatment group was the highest in each group except the normal group(P<0.05)The expression of VEGF in the sciatic nerve of each group peaked at 1,2 weeks,and began to decrease at 4 weeksSpinal cord:at three time points of 1,2,and 4 weeks,the VEGF expression of the model control group and the three treatment groups at each time point was higher than that of the normal control group(P<0.05);The expression was higher than that in the model group,the neuromobilization group and the combination treatment group(P<0.05);at the two and four-week time points,the expression in the treatment group was higher than the model group,and the expression in the combination treatment group was higher than weeks5.Blood nerve barrier leakageObserved under the microscope,the leakage degree of the model group and the three treatment groups in the 1,2,and 4 week groups was higher than that in the normal group.2.The leakage of the treatment group at two time points at 4 weeks was significantly better than that of the model group Among them,the leakage in the combined treatment group improved significantly.Conclusions:1.Jiaji electroacupuncture,nerve mobilization,and combination therapy can improve the function of sciatic nerve,increase the expression of VEGF and CD34,promote peripheral nerve and blood vessel regeneration,improve the degree of blood nerve leakage,promote the repair of peripheral nerve,and combine treatment The group is superior to nerve mobilization or spinal electroacupuncture alone2.Jiaji electroacupuncture combined with nerve mobilization to promote the regeneration of injured sciatic nerve may be related to the regeneration of peripheral nerve vessels and the improvement of blood nerve barrier leakage.
