Purpose:In this experiment,the quality of Gentiana scabra and its seed and seedling were studied in different cultivation areas of Liaoning Province,and the adaptability of the seedlings to stress stress was studied.So as providing the basis for establishing the quality evaluation of gentian and more standardized cultivation of Gentiana scabra.Materials and methods:1 materials: The experiment materials were collected from 3 different cultivated areas in Liaoning province and the origin of different base gentian 14 batches(Qingyuan County,Fushun City,Liaoning Province,Xinbin County,Fushun City,Huanren County,Benxi City),medicinal parts were its root and rhizome.The materials were identified as Gentiana scabra Bunge.By professor Yin Haibo of Liaoning University of Traditional Chinese Medicine.Gentiana seed and 2-year-old Seedling collected in Ying'e Men Zhen Ying'e Men Cun,Qingyuan County,Fushun City which all varieties are Gentiana scabra Bunge.The experimental soil samples were collected for the cultivation base corresponding to the 14 batches of medicinal materials.2 Method : 2.1 using petri dish paper germination method with PEG6000 and different concentrations of pure NaCl creating water stress and salt stress environment,on the basis of determination of seed germination rate,germination potential,germination index,etc,which the determination results by SPSS 20.0 statistical software significant difference test,to compare the difference of seed germination index between stress environment and normal growth environment.2.2 Through the literature review,6 exogenous substances that can alleviate the damage of drought stress were screened out,respectively,CaCl2,salicylic acid,chitosan,PEG,asinamide and betaine.Using the germination method of culture dish paper,PEG6000 artificially created the drought stress environment,and the external sources added six exogenous substances with different concentrations,and the results were analyzed by single factor analysis of variance analysis.2.3 Cultivation of Two-year-old Gentiana scabra seedlings in tissue culture for 5 weeks,during the normal irrigation,with pot water control method to deal with drought stress,different concentrations of chitosan solution were injected into the roots of seedlings in stress treatment group,and chlorophyll content in leaves was measured during the treatment.After 6 weeks of culture,the roots and rhizomes of seedlings were collected.After drying and crushing,the content of cleft cyclic iridoid glycosides was determined by high performance liquid chromatography(HPLC).2.4 High performance liquid chromatography(HPLC)was used to determine the contents of cyclic iridoid glycosides,that is,gentiopicroside,Swertia,Swertia,Gentiana scabra in different cultivation areas,and the contents of heavy metals and agricultural residues in the soil of different habitats.To calculate the dried rate and evaluate the quality of Gentiana gentiana.Results:1.The experimental correlation analysis showed that there was a significant negative correlation between germination rate and NaCl concentration of Gentiana gentiana seed,and the seed germination potential,germination index,seedling length and vigor index decreased with the increase of NaCl.The suitable value of salt stress was 13.15 mmol / L,the critical value was 71.06 mmol / L,and the limit value was 128.97 mmol / L during germination.When the concentration of PEG was 5,the germination rate,germination potential and germination index of Gentiana seed were increased by a small margin compared with the control group.There was no significant difference in rhizome ratio,seedling length and control group(P < 0.01).2.15%PEG drought stress significantly inhibited seed germination.6 kinds of exogenous substances could alleviate the damage caused by drought stress to a certain extent.Among them,betaine 625mg/L,chitosan 25mg/L,SNP 0.7mmol/L,salicylic acid 10mg/L,calcium chloride 5 mmol/L,spermidine 0.01mmol/L concentration were the best.3.Under drought stress,the content of gentiopicrin in the seedlings of Gentiana scabra seedlings was higher than that in the normal water control group,and the chlorophyll content in the leaves was lower than that in the normal water control group,and the content of gentiopicrin in the four concentration groups of chitosan was higher than that in the normal water control group.The content of chlorophyll was lower than that of the normal water control group and higher than that of the drought stress control group,in which the content of gentiopicrin in the 150 mg/L chitosan treatment group was basically equal to that in the drought control group(3.91%),so the optimum concentration of chitosan solution was 150 mg/L.4.The results show that the overall trend of the 4.content determination of gentiopicroside in Huanren County,Qingyuan County Xinbin county,but the results by SPSS 20.0 statistical analysis showed no significant difference(P > 0.05).Between the Qingyuan cultivation base of gentiopicroside content in base 5 and base 8 overall difference is not obvious,base 5,base the 8 content is higher.5.pesticide residues and heavy metals in the 3 Liaoning cultivated areas of copper,lead,mercury,arsenic contents of 4 kinds of heavy metals are in line with the national standard limits of heavy metals,Qingyuan County 4,5,6,8 planting base of Chinese herbal medicine did not detect mercury,all cultivated soil were not detected in cadmium element,and all base ingredients were detected the Cd element,2-phenyl phenol detection and Xinbin County Qingyuan County 2-2 base of medicinal materials,its content exceeded the EU standards(No 396/2005 twelfth).6.Qingyuan wandianzi ice town ditch cultivation base of medicinal material the drying rate was 41.22%,the highest amount of ying'emen Zhen ying village cultivation base of medicinal material drying rate is 40.51% on average,five Austin Village Huanren County,the drying rate was 38.03%,beidianzi medicinal materials drying rate of 39.67%,reduction rate of 40.95%.dry herbs Xinbin County.Conclusions: 1.The seed size of Gentiana gentiana is small,and the storage time is only about one year under normal conditions.The germination of Gentiana gentiana seeds under water stress and salt stress was studied for the first time.The results showed that the germination indexes of Gentiana gentiana seeds increased under moderate drought stress,indicating that moderate drought stress could promote seed germination,salt stress was used to artificially create salt stress environment by pure sodium chloride.The results showed that there was a significant negative correlation between the germination rate and NaCl concentration,and the germination indexes decreased significantly with the increase of sodium chloride concentration,which indicated that the seeds of Gentiana scabra mildly tolerant to drought and extremely intolerant to salt.It is necessary to prevent soil salinization,reasonable irrigation and rational fertilization to ensure the healthy and sustainable development of cultivation industry of Gentiana scabra.2.Some studies have found that some exogenous substances can alleviate the damage to plants caused by stress stress.Six exogenous substances,chitosan,salicylic acid,spermidine,betaine,calcium chloride,sodium nitroprusside,were screened by consulting the literature.The effects of six exogenous substances on seed germination under drought stress were studied.The results showed that the germination rate and other germination indexes of seeds under drought stress could be increased to a certain extent by six exogenous substances.It is proved that it is effective in alleviating drought stress.3.Studies on adaptability of seedlings to drought stress showed that drought stress inhibited chlorophyll synthesis in seedling leaves,but promoted the accumulation of gentiopicrin,a secondary metabolite of root and rhizome.Chitosan could alleviate the damage caused by drought stress.The accumulation of gentiopicrin was promoted by high concentration chitosan and drought stress.4.The quality of cultivated Gentiana scabra in Wandian Town of Qingyuan County is higher than that of other plants,but the problem of exceeding the standard of CD should be warned and the cleanliness management of land should be strengthened according to the dry rate of medicinal materials,the content of index components and the test results of heavy metal residues.
