Preliminary Study On The Efficiency Of Bombyx Batryticatus In Cosmetics Application

Bombyx batryticatus,a kind of traditional Chinese medicine,is the dried dead larvae of Bombyx mori L.(silkworm of 4?5 instars)infected by Beauveria bassiana(Bals.)Vuill.As a traditional Chinese medicine,Bombyx batryticatus has a long medicinal history.There are records in"Compendium of Materia Medica":"mix it with honey and wipe the face to whiten and freckle",and"Shen Nong's Materia Medica"contain:"eliminate black spots and make people have florid facies".These show that the Bombyx batryticatus was used as a"cosmetics"long before.Modern pharmacological studies shown that it has a wide range of pharmacological effects,such as anticonvulsant,anticoagulant,antioxidant,bacteriostatic and other effects.Cosmetics are a type of daily-use chemical products that are mainly applied topically to the skin,in order to meet the needs of consumers'appearance.With the development of economy and the improvement of people's living standards,consumers'consumption concepts tend to be rationalized,and the requirements for the safety and specific functions of cosmetics are becoming higher and higher,such as antioxidant,whitening,anti-inflammatory,bacteriostatic and so on.Therefore,giving specific effect to cosmetics is a leading direction of the development of cosmetics.In the modern society where natural ingredients are favored,traditional Chinese medicine has become a highly anticipated raw material for functional cosmetics.The Bombyx batryticatus is one of the most commonly used traditional Chinese medicine cosmetics-related medical books,but in modern cosmetics,the Bombyx batryticatus has not been used as a raw material for cosmetics.In this study,by obtaining the Bombyx batryticatus extract,evaluating its safety and screening ite efficacy,it is expected to develop the use value of the Bombyx batryticatus,and provide ideas and theoretical basis for the research and development and promotion of functional Chinese herbal cosmetics.The main research results obtained are as follows:1.Safety assessment of silkworm extractUsing water and ethanol to extract the Bombyx batryticatus,then obtained BBW and BBE.The protein content of BBW is(65.11±2.45)%,the total sugar content is(20.71±0.37)%,the flavonoid content is(0.17±0.02)%,and the ash content is(18.19±0.11)%.The flavonoid content of BBE is(2.2±0.16)%,and the ash content is(2±0)%.The IC₅₀ of BBW was 20.66 mg/m L,IC₅₀ of BBE was 329.5?g/m L.Chicken embryo chorioallantoic membrane test results show that 0.5%BBW and 0.5%BBE are not eye irritating.The above results indicate that BBW has higher cell safety and no eye irritation;BBE has lower cell safety and no eye irritation.2.Preliminary study on the efficacy of BBEThe results of DPPH free radical scavenging experiment and ABTS free radical scavenging experiment showed that BBE had the ability to scavenge DPPH free radicals and ABTS free radicals,indicating that BBE has the potential of antioxidant.The results of tyrosinase inhibition experiments showed that BBE has an inhibitory effect on tyrosinase monophenolase and tyrosinase bisphenolase.ELISA results showed that BBE could not reduce the level of inflammatory factor TNF-?secreted by LPS-stimulated RAW264.7 macrophages,and it could not increase it.But BBE could significantly reduce the level of IL-6.In order to test whether BBE has antibacterial activity,E.coli,S.aureus,P.acnes,M.furfur were selected as the test strains.The results showed that BBE had good antibacterial activity,and its inhibitory effect on S.aureus was the best,followed by P.acnes,and the inhibitory effect on E.coli was the weakest.The above results indicate that BBE has the potential to become an acne-related cosmetic raw material.3.Preliminary study on the efficacy of BBWThe results of the four in vitro antioxidant test methods showed that BBW has the ability to scavenge DPPH free radicals,ABTS free radicals,and hydroxyl free radical scavenging.As the concentration increases,the scavenging rate tends to be stable.The total antioxidant capacity of BBW determined by FRAP method was(0.37±0.01)mmol/g.The above results showed that BBW has antioxidant activity.The whitening efficacy of BBW was tested by tyrosinase inhibition experiment.The results showed that BBW had a good inhibitory effect on tyrosinase monophenolase and tyrosinase diphenolase,and had a concentration correlation,with IC₅₀ of 1.463 mg/m L and 3.02mg/m L.ELISA and q PCR showed that BBW could significantly reduce the level and expression of TNF-?and IL-6 secreted by macrophages stimulated by LPS,and had a certain concentration correlation.In vitro inhibition experiments and q PCR showed that BBW had an inhibitory effect on COX-2.Antibacterial experiment results showed that the BBW could not inhibit the growth of E.coli and S.aureus.The above results indicated that BBW could have antioxidant,whitening and anti-inflammatory effects.
