Objective:To investigate the protective influence and mechanism of Chaoyi Jiawei Danggui Wenzhong Decoction on the significant gastric mucosal injury produced by ethanol,to implement an experimental foundation for relevant study and clinical application.Methods:SD male rats were randomly divided into 6 groups: normal group,model group,sucralfate group,Jiawei Danggui Wenzhong Decoction group(high-dose group,medium dose group,low-dose group),then 8 in each group.The model was established by gavage with unblemished ethanol(0.5ml/100g),and an acute gastric mucosal injury model was formed one hour later.After flourishing modeling,the control group and model group were given normal saline(2ml/200g).So on,Suc group(2.06ml/kg),JDWD high-dose group(2.1g/kg),JDWD medium-dose group(1.05g/kg),JDWD low-dose group(0.53g/kg).After 14 days of consecutive intragastric treatment,fasting without water for 24 h,and practicing serum and stomach.Enzyme-linked immunosorbent assay was used to measure rat serum superoxide dismutase activity,malondialdehyde content,epidermal growth factor(EGF)content,prostaglandin E2(PGE2)level,and HE staining to observe the pathological changes of gastric mucosa,immune group Chemical staining method was used to observe the expression of cyclooxygenase(COX-2)protein in gastric tissue.Result:1.MDA content measurement results,the content of the JDWD groups and the Suc group were significantly decreased.Then compared with the Model group,the variation was statistically significant(P<0.05).2.SOD activity measurement results,the SOD activity of each JDWD group and the Suc group were significantly expanded,compared with the Model group,the variation was statistically significant(P<0.05).The SOD activity of the JDWD high-dose group was higher than that of the Suc group.It is statistically significant(P<0.05).3.EGF content determination results,the content of JDWD groups and Suc group improved,compared with Model group,the variation was statistically significant(P<0.05).JDWD high and medium-dose group EGF content is higher than Suc group,it is the variation was statistically significant(P<0.05).4.As a result of PGE2 content determination,the content of the JDWD group and the Suc group both increased.Compared with the Model group,the difference was statistically significant(P<0.05).PGE2 content of JDWD high and medium-dose group was higher than that of Suc group,the difference was It is statistically significant(P<0.05).5.Pathological examination results revealed that the gastric mucosal gland structure in the JDWD high-dose group was recovered intact,a small amount of superficial mucosal small blood vessels was congested,and no obvious inflammatory cell infiltration was seen.All groups of JDWD had the consequence of repairing the gastric mucosa.6.As a result of immunohistochemical staining,the expression of COX-2 in the model group was significantly higher than that in the normal group,and the expression of COX-2 in the administration group was significantly reduced,especially at high doses of JDWD.Conclusion:Chaoyi Jiawei Danggui Wenzhong Decoction can efficiently adjust the gastric mucosal injury in rats produced by ethanol.The mechanism may be associated with the reduction of malondialdehyde content.The increase of prostaglandin,epidermal growth factor,superoxide dismutase content and the diminish of COX-2 protein expression in gastric tissue.
