| Objective:Exercise,diet and orlistat are all effective ways to lose weight.The decrease of male fertility caused by obesity is a hot topic in recent studies.Three methods of fat reduction were used to intervene in male rats with obesity induced by high-fat diet,and the effects of these three methods on fat reduction,inflammation level and sperm quality in obese rats were studied.Methods:77 Experimental rats were randomly divided into 2 groups,10 in the control group(group C)and 67 in the high-fat diet group for 6 weeks.Fifty successful rats were randomly selected and divided into the high-fat control group(HF group),the high-fat exercise group(HE),the high-fat orlistat group(HO)and the high-fat diet group(CR).The high-fat control group was fed high-fat diets.The high-fat orlistat group(HO group)was given orlistat 60mg/kg daily,and the other groups were given 0.5%cmc-na solution of the corresponding volume for a total of 8 weeks.The rats in the exercise group(HE group)performed weightless swimming every afternoon for 6 days a week.The first week was adaptive exercise.The exercise group exercised for 1min on the first day and increased by 10min every day after that.After 1 week,the exercise increased to 60min.The diet group was fed a high-fat diet of less than 30 percent a day,all other things being equal.Methods serum levels of TNF-α,IL-6,Metrnl and TNF-α and IL-6 in the testicles of rats were detected by ELISA.Rt-pcr was used to detect the mRNA expression levels of the pro-inflammatory factors TNF-α,IL-6 and Metrnl.Sperm quality and malformation rate were detected by computer aided analysis(CASA)and diff-quik rapid staining,and sperm DNA integrity was detected by fluorescent staining.Results:(1)There was no significant difference in body weight and Lees index between the normal group and the high-fat group before modeling.After 6 weeks of modeling,the body weight of the high-fat diet group was 20%higher than that of the normal diet group,with an extremely significant difference PAfter modeling,the Lees index of the high-fat group was significantly higher than that of the normal group,Pcompared with group C,HC group rat body weight,Lee’s index,body fat rate,high rate of brown fat has significant difference(P<0.01),(2)8 weeks after the intervention,HE,HO group,body weight,body fat rate of CR group were lower than the HC group and has the extremely significant difference,aerobics,orlistat,dieting can effectively improve the high-fat diet obese rats.The rate of brown fat in HC group and HE group was higher than that in HO group and CR group,with significant difference P<0.01.(3)After 8 weeks of intervention,TG and ldl-c were significantly higher in HC group than in group C(Pand hdl-c was significantly lower than in group C(P<0.01).The extreme significance of FPG and TC in HO group was higher than that in other groups(P<0.01).while that in HE group and CR group was significantly lower than that in HC group(P<0.01).Compared with the HC group,the triglycerides in HE,HO and CR groups were significantly decreased by(P<0.05).High-density lipoprotein in CR group was significantly higher than that in other groups,(P<0.01).Low density lipoprotein in CR group was significantly lower than HC group(P<0.01).That aerobic exercise,orlistat,diet can regulate the status of high fat diet lipids.(4)After 8 weeks of intervention,the serum IL-6 level in the CR group was significantly lower than that in the HC group,with a significant difference of(P<0.05).The serum Metrnl level of HE group was higher than that of HC group,HO group and CR group,and there was a significant difference between HE group and CR group Metrnl level in serum of CR group was significantly lower than that of HC group(P<0.01).While IL-6 level in testicular tissue of HC group was significantly higher than that of group C.The level of TNF-α in the testis of HC group was significantly higher than that of CR group,PThe level of IL-6 in the testis of HC group was significantly higher than that of HE group and CR group,Pand significantly higher than that of HO group,(P<0.05).(5)After 8 weeks of intervention,mRNA expression levels of IL-6 and TNF-α were significantly higher in the HC group than in the C group in the well-received and adipose tissue.Metrnl mRNA expression in HC group was significantly higher than that in C group,In the adipose tissue,the expression levels of IL-6 mRNA and TNF-amRNA in the HC group were higher than those in HE,HO and CR groups,and the difference was extremely significant,pIn adipose tissue,Metrnl mRNA expression level in HC group was significantly higher than that in CR group,Metrnl mRNA expression in HE group was significantly higher than that in C,HO and CR group,with significant difference PIn muscle tissues,the expression level of TNF-α in HC group was higher than that in C group,with a significant difference of The expression level of IL-6 mrna in HE group was significantly higher than that in CR group(P<0.05)The expression level of TNF-α mRNA in the HC group was significantly higher than that in the CR group,with a significant difference of Pbut no significant difference with HE and HO.In muscle tissue,Metrnl mRNA expression level in HE group was significantly higher than that in CR and C group(nd there was a significant than that of HE group and the difference was significant Sperm density:the higher sperm density of the HO group and the CR group was significantly different from that of the C group,the HC group and than those in C group,HE group,HE group and CR group,(P<0.01)with a Pdifference.Conclusions:(1)(1)6-week high-fat diet can cause nutritional obesity in SD rats.(2)aerobic exercise,orlistat and diet had the effect of reducing weight and decreasing fat in high-fat diet rats,while the effect of aerobic exercise and diet intervention on improving fat metabolism in high-fat diet rats was more obvious.(3)aerobic exercise,orlistat and diet all have significant effects on reducing the level of inflammation in the body."aerobic exercise--increased METRNL level in muscle tissue--decreased inflammation level" may be one of the mechanisms of reducing inflammation level in aerobic exercise.Orlistat and diet can inhibit the increase of fat,thereby reducing the expression of inflammatory cytokines and improving the inflammatory state of obese rats.(4)Aerobic exercise,orlistat and diet intervention improve sperm quality by reducing fat accumulation and body inflammation. |
