| Objective:Based on XHF research basis of the Institute of Tianjin Institute of Environmental&Operational Medicine,and according to the prescription principle of anti-hypoxia and anti-fatigue,combined with the anti-hypoxia effect of traditional Chinese medicine anti-hypoxia and anti-fatigue nutrients such as taurine and CoQ10.Through the verification of the anti-hypoxia and anti-fatigue effect of the compound nutrient agent,and according to the changes of biochemical indexes in the serum or tissue of mice,the anti-hypoxia and anti-fatigue effect of the compound nutrient agent was clarified,which provides a theoretical basis for further research on the application and development of the compound nutrient agent.Methods:1.The experiment of normobaric closed hypoxia in mice.Through the establishment of a mouse model of normobaric hypoxia,0.5% sodium carboxymethyl cellulose(CMC)was used as a blank control and XHF as a positive control to observe the effects of different doses of compound nutrients on the body weight and survival time of mice under normobaric hypoxia.The activities of T-AOC,CAT,SOD,GSH-PX and MDA in serum and the activities of ATP enzyme in brain tissue were detected.2.Weight-bearing swimming test of miceThrough the weight-bearing swimming test of mice,with 0.5% CMC aqueous solution as the blank control and XHF as the positive control,the effects of low,middle and high doses of compound nutrition on the swimming time of weight-bearing mice were observed,and the contents of liver glycogen,muscle glycogen,BUN,LA,MDA and the activities of LDH and SOD in serum and tissue of mice were detected.Results:1.Anti-hypoxia effects of compound nutrients in mice(1)Effects of compound nutrients on body weight and anoxic survival time of mice.The effect of compound nutrition on the body weight of mice:there was no significant difference in the body weight of mice in the XHF positive control group,low,middle and high dose groups compared with the blank control group(P>0.05),which indicated that the compound nutrition had no adverse effect on the body weight of mice.The hypoxia survival time of mice in XHF positive group and low,middle and high dose groups was significantly longer than that in blank group(P<0.01),and the hypoxia survival time in high dose group was significantly longer than that in XHF group(P<0.05).(2)The effect of compound nutrition on serum oxidation index of mice:(1)The serum T-AOC ability of mice in middle and high dose groups was significantly higher than that in XHF positive control group(P<0.01).(2)The activity of CAT in the serum of mice in the XHF positive control group,low,middle and high dose compound nutrition groups was significantly higher than that in the blank group,and the serum CAT activity in the low,middle and high dose compound nutrition groups was still significantly higher than that in the XHF positive control group(P<0.01).(3)The activity of SOD in the serum of mice in the XHF positive control group and low-dose compound nutrition group was significantly higher than that in the blank group,and the SOD activity in the middle-dose compound nutrition group was significantly higher than that in the XHF positive control group(P<0.05).(4)The activity of GSH-PX in serum of mice in the middle dose group of compound nutrition was significantly higher than that in the positive control group of XHF(P<0.05),and the activity of GSH-PX in serum of high dose group of compound nutrition was significantly higher than that of positive control group of XHF(P<0.01).(5)The content of MDA in serum of mice in the low,middle and high dose groups was significantly lower than that in the blank group,and the content of MDA in the middle dose group was significantly lower than that in the XHF positive control group(P<0.05).(3)The effect of compound nutrition on the activity of ATPase in brain tissue of closed hypoxia mice:(1)The activity of Na+K+-ATPase in brain tissue of hypoxia mice in middle and high dose groups was significantly higher than that in blank group(P<0.01),but there was no significant difference compared with XHF group.(2)The activity of Ca2+Mg2+-ATPase in brain tissue of middle and high dose groups of compound nutrition was significantly higher than that of blank group and XHF positive control group(P<0.05).2.Anti-fatigue effects of compound nutrients in mice(1)Effect of compound nutrition on swimming time of weight-bearing swimming mice:there was no significant difference in swimming time between XHF positive control group and blank control group,but the weight-bearing swimming time of middle and high dose groups of compound nutrition group was significantly longer than that of blank group(P<0.01).(2)Effect of compound nutrition on serum indexes of weight-bearing swimming mice:(1)The activity of LDH in serum of mice in middle and high dose groups was significantly different from that in blank group and XHF positive control group(P<0.01).(2)The content of LA in serum of XHF positive group was significantly lower than that of XHF positive control group(P<0 01).The content of LA in serum of high dose compound nutrition group was significantly lower than that of XHF positive control group(P<0 01).(3)The content of BUN in serum of mice in XHF positive control group was significantly lower than that in blank group,and the content of BUN in high dose compound nutrition group was significantly lower than that in XHF positive control group(P<0.01).(3)Effects of compound nutrition on oxidation and antioxidation indexes in liver and skeletal muscle of weight-bearing swimming mice:(1)The activity of SOD in liver tissue of weight-bearing swimming mice in middle and high dose groups was significantly higher than that in blank group,and the activity of SOD in liver tissue in high dose group was significantly higher than that in XHF positive control group(P<0.01).(2)The activity of SOD in the skeletal muscle of weight-bearing swimming mice in the middle and high dose groups was significantly higher than that in the blank group(P<0.01),but there was no significant difference between each dose group and the XHF positive control group.(3)The content of MDA in liver tissue of weight-bearing swimming mice in middle and high dose groups was significantly lower than that in blank group,and the content of MDA in liver tissue of weight-bearing swimming mice in high dose group was significantly lower than that in XHF positive control group(P<0.0l).(4)The content of MDA in skeletal muscle of weight-bearing swimming mice in middle and high dose groups was significantly lower than that in blank group(P<0.01),but there was no significant difference between each dose group and XHF positive control group.(4)Effect of compound nutrition on liver glycogen and muscle glycogen in mice:there was significant difference between XHF positive control group and blank group.The storage of liver glycogen and muscle glycogen in middle and high dose groups of compound nutrition was significantly higher than that in XHF positive control group(P<0.05).The content of muscle glycogen in high dose compound nutrition group was significantly higher than that in XHF positive control group(P<0.05).Conclusion:The main results are as follows:(1)Compound nutrient can prolong the survival time of mice under normobaric hypoxia and has anti-hypoxia effect;it can reduce the production of free radicals,alleviate the disturbance of cell energy metabolism,protect the cell membrane damage caused by lipid peroxidation,improve the oxidative damage induced by hypoxia,increase the activity of ATP enzyme in the brain tissue of mice under normobaric hypoxia,and protect the brain tissue under hypoxia.(2)Compound nutrition can prolong the swimming time of mice,increase the reserve ability of liver glycogen and muscle glycogen,reduce the contents of LA,BUN and MDA in serum,liver and skeletal muscle,enhance the activity of LDH and SOD in serum,liver and skeletal muscle,reduce muscle injury and enhance the anti-fatigue ability of mice.(3)After taurine and CoQ10 were added on the basis of XHF,the overall anti-hypoxia and anti-fatigue effects of the compound nutrient agent were better than those of XHF,showing a good synergistic effect. |
