| Cordyceps militaris is an important edible fungus for both food and medicine,and its albino strains have also received great attention from researchers.In this experiment,stable genetic albino strains of Cordyceps militaris were used as the research objects to study the differences in biological characteristics,antioxidant enzymes,and main metabolite contents between albino strains and normal strains.Scientific evaluation of albino strains is of great significance for exploring the albino mechanism of Cordyceps militaris.The test results are as follows:1.In this experiment,it was verified by ITS sequencing that it was a Cordyceps militaris strain,and the sequence comparison similarity rate reached 99%.2.Experiment Within the scope of this experiment,the colony morphology and growth rate of Cordyceps militaris albino strains and normal strains were significantly different.The colony of the albino strain was white,without macrohorn change,and the rhythm ring was not obvious;the colony of the normal strain was orange-yellow,with no aerial hyphae leggy and obvious rhythm ring.Compared with the control,the mycelial growth rate of the albino strain showed the characteristics of fast-slow-fast,and the mycelial growth rate was significantly faster.Under the same culture conditions,the albino strain expanded faster,and the fresh weight of the mycelium was 168.6% of the control.3.By annotating the differential genes,a total of 2068 differential genes were annotated in the normal strain B3 of Cordyceps militaris and the albino strain BC,1483 were annotated in GO,and 546 were annotated in KEGG,including 917 up-regulated genes and 1151down-regulated genes.indivual.4.Under the conditions of this experiment,the activities of antioxidant enzymes SOD,POD,MDHAR and APX of the albino strain were significantly increased compared with the control.The staining of reactive oxygen species was observed by laser confocal microscope,and it was found that the tips of the two strains did not contain reactive oxygen species,which were considered to be the tip of the mycelium as the growth point,with strong vitality and strong scavenging ability of reactive oxygen species.Transcriptome sequencing analysis found that isocitrate dehydrogenase and glutathione S-transferase were up-regulated in glutathione metabolism,and glutathione hydrolase and glutamate-cysteine ??ligase catalytic subunit were down-regulated.Among them,the increase of isocitrate dehydrogenase content promoted the generation of GSH in the glutathione cycle,and increased the enzymatic activity of the antioxidant enzyme system of the albino strain.5.Cordycepin and adenosine,the main metabolites of the albino strain,were significantly higher than those of the normal strain.The content of cordycepin in the albino strain was 9.8565±0.1284mg/ml,an increase of 22.7%,and the content of adenosine was6.7499±0.1834mg/ml,an increase of 22.7%.22.9%.Through transcriptome data analysis,it was found that the synergistic effect of up-regulation of adenosine succinate synthase(CM_6725)and down-regulation of adenosine hydrolase(CM_2903,CM_6130)promoted the accumulation of cordycepin and adenosine.6.Through transcriptome data analysis,the metabolic pathways related to pigment synthesis and accumulation were screened.Among them,8 differential genes were annotated in tyrosine metabolism,and the expression of tyrosinase gene(CM_1704)was significantly down-regulated,which made the albino strains.The accumulation of melanin is reduced,which promotes the occurrence of the whitening process.Reliable transcriptomic data determined by real-time fluorescence quantification. |
