| Among common pets,the incidence of canine tumors is high.In recent years,the incidence of canine female reproductive system tumors has been on the rise.The canine female reproductive system has a complex tissue structure and is prone to various histological changes,which increases the difficulty of pathological diagnosis for pathologists.In this study,the expression of PCK,CK8/18,E-cad,CAM5.2,HBME-1,CD117,Melan-A,Vim,α-SMA,S-100,NSE,Inhibin-α and other related immune markers in normal canine female reproductive system tissues was studied,combined with histology to differentiate and diagnose canine female reproductive system tumors and tumor-like lesions.In this way,an immunohistochemical panel for the differential diagnosis of canine female reproductive system tumors and tumor-like lesions was screened.It provides methods for accurate diagnosis of female reproductive system tumors and tumor-like lesions in dogs,accumulates relevant clinical data,and provides reference for the prevention and treatment of female reproductive system tumors and tumor-like lesions in small animals.This study found that the ovarian surface epithelium of clinically healthy dogs was positive for PCK,CK8/18,CAM5.2,HBME-1,Vim and Melan-A,and cortical tubules were positive for CK8/18,CAM5.2,HBME-1,Vim and Melan-A;follicular granulosa cells and granulosa cell cords were positive for Vim,Melan-A and Inhibin-α;the luteal cells were positive for Vim and Melan-A;epithelial cells of the rete ovarii were positive for PCK,CAM5.2,HBME-1 and Vim.The IHC results of the mucosal and glandular epithelium of clinically healthy endometrium showed positive for CAM5.2 and E-cad,positive for PCK in the mucosal epithelium,and negative for PCK in the glandular epithelium.Clinically healthy canine vaginal stratified squamous epithelium was positive for PCK,positive forα-SMA and NSE in the muscle layer,negative for S-100 and CD117,and Masson staining showed blue collagen fibers in connective tissue.This trial retrospectively studied 53 canine female reproductive system tumors and tumor-like lesions based on the IHC characteristics of clinically healthy canine female reproductive system tissues.The results of this study show that ovarian epithelial tumors can use PCK,CK8/18,CAM5.2,HBME-1 positive markers,Melan-A and Inhibin-α can be used as positive markers for ovarian sex cord-stromal tumors,only CD117 positive expression is dysgerminoma.The identification of ovarian cysts is mainly based on their histological location and the IHC expression characteristics of the cells lining the cysts.SES cysts are located in the ovarian cortex,and cystic rete ovarii are located in the ovarian medulla,and the cystic lining cells of both are positive for PCK,CK8/18,CAM5.2 and HBME-1;the cystic lining cells of follicular cysts are granulosa cells,namely Melan-A and Inhibin-α were positive;the cyst wall of cystic corpus luteum was positive for MelanA;the surrounding cavity of paraovarian cyst was positive for α-SMA.Masson staining of polyps in utero-vaginal tumor-like lesions showed blue collagen fibers with regular arrangement.Smooth muscle-derived tumors were positive for α-SMA;fibroids and fibrosarcomas showed abundant blue collagen fibers by Masson staining,which were mainly distinguished from polyps by the interweaving of collagen fibers in fibroids and fibrosarcomas.This study shows that by studying the expression rules of relevant immune markers in normal canine female reproductive system tissues,an immunohistochemical panel that can be used for accurate diagnosis of canine female reproductive system tumors and tumor-like lesions has been preliminarily screened. |
