In this thesis,the first round of ARTP mutation breeding technology was used to obtain Aa66 as the starting strain,and then the MPMS technology was used to carry out further mutation breeding work to obtain 315 mutant strains;29 mutant strains in the initial screening produced significant antagonistic reactions with the starting strain Aa66;subsequent re-screening tests were conducted,including the comparison of the growth rate of the mutant strain and the starting strain,and the comparison of the three extracellular enzyme activities(carboxymethyl cellulase enzyme activity,laccase enzyme activity,and hemicellulase enzyme activity).In the follow-up screening test,including the comparison of the mycelial growth rate of the mutagenic strain and the starting strain,the comparison of the three extracellular enzyme activities(carboxymethyl cellulase enzyme activity,laccase enzyme activity,hemicellulase enzyme activity),and the comparison of biological efficiency,it was determined that the traits of the mutagenic strain Y9,Y16,Y37,Y50,Y180 were better than the starting strain Aa66,among which the mutagenic strain Y9 had the best traits,the mycelial growth rate increased by 11.65% and the three enzyme activities increased by 21.65%compared with the starting strain.The three enzyme activities increased by 21.62%,20.65%and 6.54%,respectively,and the biological efficiency was 115.34%,which was 8.43% higher than that of the starting strain.The authenticity of the five excellent mutagenic strains Y9,Y16,Y37,Y50,Y180 and the starting strain were identified,and the results showed that the genetic coefficients of variation between the five mutagenic strains and the starting strain ranged from 0.475 to 0.833,which proved that there were different genetic differences between the five mutagenic strains and the starting strain.In the preparation process of liquid strain-original seed of grain,by studying the effect of different grains on the nutritional growth of Auricularia auricula,comparing the inoculation of liquid strain and PDA strain,the growth rate of Auricularia auricula mycelium on five kinds of grain medium,it was determined that wheat was the most suitable grain as the medium,and the growth rate of liquid strain was the fastest,with a growth rate of 5.62mm/d;the best formula of original seed medium was 88% of wheat grain,10% of mixed wood chips and 2% of lime.To study the effect of different additions on the nutritional growth of Auricularia auricula,it was determined that 15 m L of liquid strain was the optimal amount to be added to the "wheat grain + 10% mixed wood chips" medium,and the mycelial growth rate reached 6.53 mm/d.To study the effect of different seed ages on the nutritional growth of Auricularia auricula,it was determined that 15 m L of liquid strain was the optimal amount to be added to the "wheat grain + 10% mixed wood chips" medium,and the mycelial growth rate reached 6.53 mm/d.The results showed that 96 hours was the optimal seeding age,and the mycelial growth rate was 6.64 mm/d.The effect of different seeding ages on the nutritional growth of Auricularia auricula was investigated.In the application of new biological agents for Auricularia auricula cultivation,by comparing the effects of different dilutions of crustacean stimulant on the nutritional and reproductive growth of Auricularia auricula,it was determined that the ratio of crustacean stimulant was 1:1000 times,the fastest mycelial growth of Auricularia auricula reached 5.36mm/d,which was 8.28% higher than the control;the ratio of 1:1500 when crustacean stimulant was sprayed on Auricularia auricula,the biological The biological efficiency of Auricularia auricula was the highest at 126.58%,which was 11.39% higher than that of CK.Comparing the contents of polysaccharides,dietary fiber,protein and total flavonoids of the substrates of the five mutagenic strains Y9,Y16,Y37,Y50,Y180 and the starting strain and the commercially available Auricularia auricula,the results showed that the four components of the substrates of the two mutagenic strains Y9 and Y37 were higher than those of the starting strain and the commercially available Auricularia auricula,strain Y50 only had lower dietary fiber content than the starting strain,strain Y16 only had lower protein content than the starting strain,and strain Y180 only had lower total flavonoids than the starting strain.Therefore,it was concluded that the relevant components of the substrates of the five mutant strains Y9,Y16,Y37,Y50,Y180 and the starting strain Aa66 were better than the commercially available Auricularia auricula under the cultivation medium.The content of all four components of the substrates of strain Y9 was higher than that of the starting strain Aa66 and the commercially available fungus,and the content was the highest,and strain Y9 was determined to be the optimal strain.
