Typhonium flagelliforme(Lodd.)Blume,also known as Feng Goushu,Tu tianqi,is a plant of Typhonium,It is mainly distributed in Guangxi,Guangdong,Jiangxi,Anhui and other places.It grows in field ditches,rivers and lakes wetlands.Dry tubers are used as medicine,while external use mainly focuses on bleeding and swelling on the skin surface and insect bites.In recent years,with the deepening of relevant research,it has also been found that it has certain anti-tumor and anti leukemia effects on lung cancer,breast cancer,etc.Through literature research and the previous work of the research group,it has been shown that its extract has a good anti-tumor effect.On this basis,in order to further study the anti-tumor components and effects of Rhizoma Typhonii Flagelliformis,this study continued to isolate and identify the components of Rhizoma Typhonii Flagelliformis.In this paper,the separation and purification work is mainly carried out by a variety of column chromatography and high performance liquid chromatography;The structure was determined by nuclear magnetic resonance spectroscopy,high resolution mass spectrometry and physical and chemical methods.In this paper,32 compounds were obtained,including 11 alkaloids,2 phenolic acids,4 phenylpropanoids,3 glycosides,5 organic acids and 7 fatty hydrocarbonsand their derivatives.Compounds 2~6,9~13,16~17,19~21,24~27 and 29~32 were isolated from Rhizoma Typhonii Flagelliformis for the first time,and compounds 13*and 21*were new compounds.The names of the compounds are as follows:adenosine(1),N2-(1-methox ycarbonylethyl)guanosine(2),protocatechuic acid(3),phenylethylamine(4),(S)-2-hydroxy-3-(4′-hydroxyphenyl)propanoic acid(5),gentisicacid(6),L-tryptop han(7),(-)-(3S)-1,2,3,4-tetrahydro-β-carboline-3-carboxylic acid(8),(3S,6S)-3-ph enylmethyl-6-amidocarboxymethyl-piperazine-2,5-dione,cis-cyclo(Asn-Phe)(9),(-)-(1S,3S)-1-methyl-1,2,3,4-tetrahydro-β-carboline-3-carboxylic acid(10),(S)-2-h ydroxy-3-phenylpropionic acid(11),nicotinamide(12),prolylglycylprolyltryp tophylvaline(13*),(+)-isolariciresinol(14),pinoresinol(15),stagonoculiazin(16),1-O-hexadecanoyl-3-O-β-D-galactopyranosyl-glycerol(17),linolenic a cid(18),1-glyceryl linolenate(19),1-linoleoylglycerol(20),2,3-dihydroxypro pyl-13-phenyltridecanoate(21*),benzenetridecanoic acid(22),methyl linoleate(23),(E)-9-methyl elaidate(24),neoengleromycin(25),16-hydroxyhexadec-13-enoic acid(26),(2S)-1-O-(9Z,12Z,15Z-octadecatrienoyl)-2-O-(10Z,13Z-hexadecadi enoyl)-3-O-β-D-galactopyranosyl glycerol(27),linoleic acid(28),(S)-2-(palmit oyloxy)-3-(((2R,3R,4S,5R,6R)-3,4,5-trihydroxy-6-(hydroxymethyl)tetrahydro-2H-pyr an-2-yl)oxy)propyl(9Z,12Z)-octadeca-9,12-dienoate(29),1,3-dihydroxypropan-2-y l dotriacontanoate(30),10,13-octadecadienoic acid(31),(8-hydroxymethylen)-tri eicosanyl acetate(32).In this paper,the compounds isolated and identified from different extraction parts of the water extract of Rhizoma Typhonii Flagelliformis were screened for preliminary antitumor activity.Taking human lung cancer cell line(A549)as the research object,CCK-8 method was used for the experiment.Results:Compounds16,17 and 21 showed a certain inhibitory effect on human lung cancer cell line(A549),with IC50of 49.84μM,38.28μM,25.63μM.In addition,compounds 16,17and 21 mainly come from small polar sites with similar polarity,so the compounds in this polar site may be the material basis for Rhizoma Typhonii Flagelliformis to exert its anti-tumor activity.
