As a famous and precious traditional Chinese medicine,dragon’s blood has been used in treating bruises,injuries and abdominal pain.Modern pharmacological studies on dragon’s blood indicate that flavonoids are major ingredients and have effects of anti-inflammation,alleviating pain,anti-diarrhea,hemostasis,anti-ulceration,antimicrobial activity and cytotoxicity.To solve the contradiction between the increasing demand for dragon’s blood and its limited supply,this study investigated on chemical constituents of artificially induced dragon’s blood and their biological activity to supply scientific base to see if artificially induced dragon’s blood can be used instead of natural wild dragon’s blood.The crude extract of artificially induced dragon’s blood was chromatographed by various of chromatographic separation techniques(thin layer chromatography,silica gel column chromatography,Sephadex LH-20,revised phase C18 silica gel column chromatography,and preparative high-performance liquid chromatography)leading to the isolation and identification of twenty-eight compounds by spectroscopic methods(one-dimensional and two-dimensional nuclear magnetic resonance spectroscopy,high resolution mass spectroscopy,electro spray ionization mass spectroscopy,infrared radiation spectroscopy,ultraviolet spectroscopy):10-methoxy-11-hydroxydracaenone(1),4-methoxy-10,11-dihydroxydracaenone(2),1-methoxy-2-methyl-10,11-dihydroxydracaen-one(3),10,11-dihydroxydracaenone(4),10-hydroxy-11-methoxydracaenone(5),7,10-dihydroxy-1 1-methoxydracaenone(6),comboquartermer(7),cochinchinenin(8),cochinchinenine I(9),7,4’-dihydroxy-8-mthoxyflavone(10),7,4’-dihydroxyflavone(11),sappanchalcone(12),5-hydroxy-7-methoxy-1-(4’-hydroxybenzyl)-4-chromanone(13),(E)-3-(3,4-dihydroxybenzylidene)-7-hydroxy-chroman-4-one(14),3-deoxysappanchalcone(15),3’-methoxy-socotin-4’-ol(16),socotin-4’-ol(17),cochinchinenine G(18),(2S,γR)-8-methylsocotin-4’-ol(19),4’-methoxy-8-methylsocotin-3’-ol(20),(2S,γR)-3’-methoxy-8-methylsocotin-4’-ol(21),homoisosocotin-4’-ol(22),diospyrosin(23),syringaresinol(24),pinoresinol(25),medioresinol(26),bis(2-ethylhexyl)phthalate(27),methyl p-hydroxybenzoate(28).Among them,1-3,7 and 9 are new compounds,and 10 is a new natural product.The result show that the chemical constituents of artificially induced dragon’s blood are same on the whole with those from natural dragon’s blood.The new compounds with different substituting groups or formed by cyclization and/or polymerization of chalcones.Some compounds were evaluated for their inhibitory of pathogenic microbes of human and plant pathogenic fungi,and for their cytotoxic activities against five human cancer cell lines.The bioassay showed that 7,4’-dihydroxy-8-mthoxyflavone(10)and 7,4’-dihydroxyflavone(11)inhibited methicillin-resistant Staphylococcus aureus,and 16,17 and 22 exhibited significant cytotoxic activities against five human cancer cell lines with IC50 values ranged from 1.4 to 28μM.3-Deoxysappanchalcone(15)displayed moderate cytotoxicity.Base on above results,artificially induced dragon’s blood has the potential to instead natural dragon’s blood,which need further investigation on its pharmacological activity.
