Species of D.angustifolia belongs to a member of Dracaena of Liliaceae Traditionally,D.angustifolia was used as a medicinal plant in the people of Li-nationality in Hainan Island for a long time.Modern pharmacological studies demonstrated that the chemical constituents of D.angustifolia were available for blood stasis,hemostasia,acesodyne and myogenic healing.At present,there are a few researches on the volatile oil compositions of the leaves from D.angustifolia in Hainan.However,few reports of literature is available on the exact chemical constituents of D.angustifolia stems and their bioactivities.Based on that,further explorations for the chemical compositions with biological activities of D angustifolia stems were performed.In this paper,the chemical constituents and biological activity of ethyl acetate extracts of D.angustifolia were systematically studied,while the biological activity of the identified compound was also evaluatedAfter extracting the ethanol extracts from the stems of D.angustifolia with petroleum ether and ethyl acetate,extracts with different polarities were obtained The antioxidant activity was tested,and it was found that the ethyl acetate part had good antioxidant activity.Further the sample was separated by column chromatography and HPLC,attaining totally 31 compounds(1-31).they were identified as ten flavonoids derivatives tazettone H(1),(2R)-3',7-dihydroxy-5',5-dimethoxy-8-methlflavone(2),(2R)-4',7-dihydroxy-3',5-dimethoxy-8-methlflavone(3),(2R)-4',7-dihydroxy-5-methoxy-8-methylflavanone(4),4',5-dihydroxy-7-methoxy-8-methylflavan(5),4'-hydroxy-5,7-dimethoxy-8-methylflavan(6),(3S)-3,5,7-trihydroxy-4'-methoxy homoi soflavonone(7),5,7-dihydroxy-6-methyl-3-(4'-hydroxybenzyl)chroman-4-one(8),desmethylisoophiopogonone B(9),5,7-dihydroxy-3-(4'-hydroxybenzyl)chromone(10),five lignans as angudracanolide A(11),(+)-syringaresinol(12),de-4'-O-methyl-yangambin(13),(+)-isolariciresinol(14),zhebeiresinol(15),three terpenoids as 3-hydroxy-4,7-megastigmadien-9-one(16),isololiolide(17),euscaphic acid(18),three alkaloids as aurantiamide acetate(19),trans-N-p-coumaroyltyramine(20),1-H-indole-3-aldehyde(21),a sterol?-sitosterol(22),seven aromatic derivatives as ?-hydroxypropiovanillone(23),3-hydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-propanone(24),erythro-7R,8S-7-O-ethylguaiacyl glycerol(25),caserinone(26),4,6-dichloro-5-methylbenzene-1,3-diol(27),4-hydroxy-3,5-dimethoxy-benzaldehyde(28),dibutyl terephthalate(29),and other categories as D-mannitol(30)and stearic acid(31).Among them,compounds 1-4 and 11 were new compounds while compounds 7,9,10,14-17,19,23,24,26-28,30 were obtained from Dracaena for the first time.The identified compounds were tested for anti-inflammatory activity,antioxidant activity,and ?-glucosidase inhibitory activity.The results indicated compounds 2,9,10 showed obvious anti-inflamematory activity with IC50 values in the range of 33 to 61 ?M,contrast to positive control of dexamethasone with an IC50 value of 37 ?M.Compounds 3,5,10-14,and 28 exhibited evident antioxidant activity with IC50 values in the range of 0.15 to 0.35 mM,relative to positive control of trolox with a IC50 value of 0.45 mM.Compared with the positive control acarbose(4.07 mM),compounds 5,6,8,10,20,25,and 27 showed better?-glucosidase inhibitory activity.Among them,compounds 20 and 25 showed significant ?-glucosidase inhibitory activity,with IC50 values of 1.99 ?M and 0.4 mM,respectively,and the IC50 values of the remaining compounds were 0.24 to 0.78 mM.The above results indicate that D.angustifolia has good potential in the development of anti-inflammatory drugs and hypoglycemic drugs.
