In this experiment, the relationship between clinical aspect and microcirculatory disorder during experimental coliform clinical mastitis was studied. The significance of regulating microcirculation, as an important principle, in the treatment of clinical mastitis was also discussed.In the first part of the experiment, 107cfu/gland/30Kg(body weight) of E coli.(O128:K99,F41) culture were infused through the teat canal into one half of the mammary gland in 7 dairy goats. Within 4 hours, all goats expressed significant clinical signs. Milk cultivation showed that the clearance of E. coli from challenged glands increased along with promotion of the degree of inflammatory present. Excessive secretion of TNF-a and IL-6 were detected lasting for at least 72 hours after challenge of E coli., and a distinct exacerbation of hemorheology (the whole blood viscosity at shear rates of 5S-1 and 200S-1 and it affecting factors such as plasma viscosity, plasma fibrinogen concentration, whole blood reduced viscosity, and erythrocyte aggregation were significantly higher than pre-challenged) were detected during the whole process, indicating that microcirculatory disorder contributed to the continuance of clinical signs. Excessive increase of intracellular free calcium in red blood cell was also detected and was considered to be one of the main reasons causing exacerbation of hemorheology. Possible existence of multi-cell intracellular free calcium excessive increase was suspected to be the common mechanism of various aspect of microcirculatory disorder.In the second part, a new medicine named with Fu Fang Da Huang Guan Zhu Ji was developed. The new medicine was an none antibiotics intra-mammarily infused oil based on Rhubarb----a herbal microcirculatory regulator. New medicine didn't show antimicrobial activity against E coli, streptococcus agalactiae, and Staphytococcus aureus.In the third part, 24 experimentally induced clinical mastitis model were randomly divided into 3 groups assigned to receive one of three treatments: none treatment, antibiotics intra-mammarily infusion, or Fu Fang Da Huang Guan Zhu Ji intra-mammarily infusion. The result showed that Fu Fang Dahuang Guan Zhu Ji significantly decreased the concentration of TNF-a and IL-6, it also significantly decreased the whole blood viscosity of both shear rates by decreasing the concentration of fibrinogen and increasing erythrocyte deformability. All these effects of Fu Fang Dahuang Guan Zhu Ji on microcirculation may base on its regulatory effect of intracellular free calcium, while antibiotics didn't show those effects.In the forth part of the experiment, Fu Fang Da Huang Guan Zhu Ji was used against bovine clinical mastitis and showed no significant difference with antibiotic treatment in curative effect, indicating that Fu Fang Da Huang Guan Zhu Ji can be used as a replacer of antibiotics. Fu Fang Da Huang Guan Zhu Ji 20ml/3 times/quarter was recommended in treatment against bovine clinical mastitis.
