Essential hypertension (EH) is a kind of disease that seriously affects the health and living quality of human being. The morbidity of essential hypertension is on the increase in recent years. Some syndromes such as left ventricles hypertrophy and hemorrhage of the brain are threatening the health of people greatly and resulting in the decrease of their living qualities.As EH is dependent on many factors including heredity, environment, biology, and its occurrence is related to the turbulence of physical metabolization, the pathological changes and the functional maladjustment involved in many organs and systems, only lowering the blood pressure could not completely protect the target organs and tissues, and reverse their damages. Therefore, it is important to seek a effective therapy with the assistence of traditional Chinese medicines and exert their integral advantages. Some correponding studies show that traditional Chinese medicines have definitely curative effects as evidenced by improving clinic symptoms and physical metabolization, incresing living qualities, decreasing syndromes of cordis and cerebrovascular systems, and the damage of target organs. As a result, traditional Chinese medicines will present a tremendous development potentials and have a good future.Considering that the pathomechanisms of essential hypertension are very complex, professor Gai guo-zhong thinks that the key pathomechanism is disharmony of Qi and Xue, and counterflow of liver Qi, Tan Zhuo Yong Sai are the most important factors leading to the disease. It has been proved that the curative effect of Semen Raphani is reliabe to decreasing blood pressure of the patient owing to its function of JiangQiHuaTan and JongFuXieZhuo. To explore the effects and the hypotensive mechanisms of Semen Raphani, we extracted the soluble Semen Raphani alkaloids, and used the spontaneous hypertensive rats (SHR) as research object. The observations include the effect on blood pressure, serum nitric oxide (NO) content, cardiac nitric oxide synthase (NOS) activity, plasm angiotension- II (Ang II) and endothelin (ET) content, the ratio of ET and NO (ET/NO), serum malondialdehyde (MDA) content, serum superoxide dismutase (SOD) activity, the expression of endothelial nitricoxide synthase (eNOS) in aorta, the expression of eNOSmRNA in myocardium, ventricle remodeling and nephritic vascular remodeling of SHR.The results show:The soluble Semen Raphani alkaloids can decrease blood pressure of SHR;It can regulate the content of vascular active matter such as nitric oxide, angiotension-II, endothelin, increasing the NO content and decreasing the AngII and ET content;It has antioxidation function via reducing serum malondialdehyde content and enhancing serum superoxide dismutase activity;It can increase activity of nitricoxide synthase and the expression of endothelial nitricoxide synthase mRNA;(5) It can reform ventricle remodeling and nephritic vascular remodeling of rat.This research discussed various mechanisms of the soluble Semen Raphani alkaloids, ofered some objective basis for understanding and using Semen Raphani.
