Objectve:To invesigate the influence and mechanism of Xuefu zhuyu tang on blood stasis model of vascular smooth muscle cells(VSMC) proliferation in experimental atherosclerosis(AS) rabbits.Methods:30 male white big ear rabbits were randomly divided into 3 groups:normal group,model group,Xuefu zhuyu tong(XFZYT) group. Besides the normal group,each of the rabbits was fed high cholesterol diet for 10 weeks.The changes of bulbar conjunctival microcirculation were detected at 10th weekend.The blood was drawn to determine serum lipid and hemorheoloy;the level of platelet-derived growth factor(PDGF) in serum was detected by enzyme linked immunosorbent assay(ELISA);the expression of smooth muscleα-actin(SMα-actin) and proliferating cell nuclear antigen(PCNA) were detected by immunohistochemical assay;the expression of extracellular regulated kinase mRNA(ERK1/2mRNA) was determined by situ hybridization.The samples of arteries were taken for the histopathological changes of the aorta artery analysis.Results:(1)Compared with the nomal group,the model group's score values of the morpha,the flow profile and the integration in BCM are obviously increased;serum lipid and PDGF increased also;blood rheologie characteistics became worse;the expression of SM a-actin decreased,but the expression of PCNA and ERK1/2mRNA increased;The intimal hyperplasia could be found.A lot of lipid foam cells migrated to the regions of intima.The thickness of hyperplastic intima,the ratio of intima and media thickness,the ratio of intima and media area increased.(2)Compared with the model group,XFZYT group's score values of the morpha,the flow profile and the integration decreased;levels of TC,LDL-c in serum,whole blood viscosity of high share rate,whole blood viscosity of low share rate,plasma viscosity and hematocrit decreased dramatically;levels of PDGF in serum decreased also;the expression of SMα-actin increased,but the expression of PCNA and ERK1mRNA decreased;The thickness of hyperplastic intima, the ratio of intima and media thickness,the ratio of intima and media area depressed.Conclusion:serum lipid increased,blood rheologie characteistics became worse,serum PDGF increased,expression of PCNA and ERK1/2mRNA increased,expression of SM a-actin decreased,phenotypic switching of VSMC and microcirculation dysfunction.Perhaps these are the formative mechanism of blood stasis model of VSMC proliferation in experimental AS.XFZYT can improve microcirculation,regulate lipid, improve hemorheoloy,reduse serum PDGF,raise expression of SMα-actin,reduse expression of PCNA and ERK1mRNA.XFZYT can prevent the development of the AS and VSMC proliferation.
