Gardenia jasminoides var. radicans Makino, also named Gardenia jasminoides var.grandiflora, is the dried fruit of Gardenia jasminoides Ellis var.grandiflora nakai, which is a forma of Gardenia jasminoides Ellis. At present, it was mainly used externally as a resolutive in the treatment of sprains and bruises or as the raw materials for extracting gardenia yellow. It has been reported that content of the main components in Gardenia jasminoides var. radicans Makino was obviously higher than that in Gardenia jasminoides Ellis. Besides the size of Gardenia jasminoides var. radicans Makino is large, the yield is high and Gardenia jasminoides Ellis var.grandiflora nakai is easy to cultivate and manage. Thus, it was suggested Gardenia jasminoides var. radicans Makino could be used as the alternative of Gardenia jasminoides Ellis and it should be developed and utilized in order to take full advantage of the natural resources.In present study, Gardenia jasminoides var.radicans Makino was used to extract gardenia yellow, then the macroporous resin was used to purify the extracted gardenia yellow. Through the selection of the resin, the sample solution concentration, the adsorption flow rate, desorption agent, desorption flow rate and so on, the optimum condition for the purification of gardenia yellow was finally obtained. And the optimum condition was as follows:HPD722was chosen to purify gardenia yellow. The crude gardenia yellow solution with concentration of15mg/mL was loaded in column packed with HPD722resin at the flow rate of1.0mL/min, and the adsorbate-laden column was gradiently washed with800mL water,600mL15%ethanol water solution respectively at the speed of2.5mL/min, then eluted with200mL80%ethanol water solution at the speed of3.5mL/min. Under the optimun condition, the colority of the product we obtained were up to300.To take full advantage of gardenia resources, the silica gel chromatography with the dichloroethane ethanol system as mobile phase was used to separate geniposide from the waste liquid of gardenia yellow purification. Finally the purity of the geniposide product we obtained was up to90.23%and the yield was34.0%. In this study, the simultaneous extraction of gardenia yellow and geniposide from Gardenia jasminoides var. radicans Makino with ultrasonic method was also researched. Firstly, the appropriate levels of the factors affecting the extraction effect were determined by the single-factor test. Then the response surface experiment was designed with Box-Behnken central composite design method by Design-Expert8.05software. It was revealed that ethanol concentration had extremely remarkable effect on the extraction of gardenia yellow and geniposide, solid-liquid ratio had highly remarkable effect while extraction time had little effect. The optimum extraction condition was obtained by response surface methodology as follows:ethanol concentration44%, extraction time29.60min and solid-liquid ratio1:26.85. Under the condition, the actual extraction rates of gardenia yellow and geniposide were45.78mg/g and111.65mg/g, respectively. Compared with the predicted values, the differences were1.86%and1.18%, respectively. Compared with solvent reflux method, the extraction rate of geniposide has little difference while the rate of gardenia yellow increased by22.56%.
