Extraction And Quality Analysis Of Gardenia Yellow

The fruits of Gardenia(Gardenia jasminoides)are applied in China as both traditional Chinese medicine and edible foods,showing multiple functions such as purging fire,heat dampness,cooling blood detoxification etc.Modern research revealed geniposide,crocins,chlorogenic acid esters,pectin and oil as functional ingredients within Gardenia fruits.Among them,geniposide was the main component with detoxification effect,and the crocins extracted from Gardenia(gardenia yellow)had the functions of anti-oxidative damage,prevention of cardiovascular and cerebrovascular diseases and other pharmacological effects as that of saffron.The recent applications of Gardenia fruits were introduced in this thesis.Two commercial Gardenia fruits were applied in general with different application.Gardenia(Gardenia jasminoides Ellis)were adopted as herbal TCM,while water Gardenia(Gardenia jasminoides var.Radicans)were used as dyeing pigments.There existed few reports on the quality evaluation of water gardenia.In this paper,both geniposide(1)and crocin-?(2)were determined to evaluate quality of water Gardenia using a HPLC dual-wavelength method.The chromatographic conditions were as follows:a Cosmosil 5 C18-AR-? column(250 mm × 4.6 mm,5 ?m)was applied;the mobile phase consisted of a gradient of methanol and 0.1%acetic acid;the flow rate was 1.0 ml,the detector wavelength were 238 nm(for 1)and 440 nm(for 2).The results showed that both components were well separated.The linear ranges of geniposide and crocin-I were 50.16-501.6 ?g/mL(r = 0.9999),and 10.13-101.3 ?g/mL(r = 0.9997).The recovery of samples was 97.77%and 97.39%,respectively.In addition,the color value of fresh fruits was also assayed either.On the basis of these quality analysis methods,the extraction method of Gardenia yellow from water gardenia was then exploited.Through the extraction condition tests,taking into account the commercial production costs,the optimal extraction process was suggested as follows:water was chosen as extraction solvent,extracted 2 times at 80? and each for 60 min.The method has the advantages of low cost,high efficiency,simple and convenient operation,which could give some suggestions for industrial extraction of Gardenia Yellow.
