In view of studying the probable way of the explant regeneration and suitable condition of culture for tissue culture and rapid propagation in Cardiocrinum giganteum, a series of experiments were established in which the scales were employed. By means of orthogonal experiment design, analysis of variance and multiple comparisons, most favorable conditions for plant regeneration were found including basic media, concentrations and types of plant growth regulators and the dosage of activated charcoal. The results are as follows:(1) As inoculation explant, the middle scales of Cardiocrinum giganteum were better than other parts of the scales, in which the plant cells divided and formed the callus more easily. Explants isolated in March and September had higher callus formation rate and lower contamination rate.(2) Primary culture: MS+BA2.0mg/L+KT1.0mg/L+NAA0.1mg/L+IBA0.3mg/L was the best initiation medium in which the rate of initiation was 85.96%, and large numbers of buds could be differentiated.(3) Subculture: The best subculture medium was MS+BA1.5mg/L+NAA0.3 mg/L, where the subcultured plantlets grew well and after 30-35 days of subculture, the multiplication rate was 6.21; the differentiation rate of the shoots longer than 2cm was 54.80%. Subculture generation had effect on propagation and differentiation of shoots and it had better not exceed the eighth generation.(4) Rooting culture: 1/2 MS+IBA2.0mg/L+AC0.1% was the best rooting medium, in which the rooting rate was 96.87%, the number of roots was 7.2, the average length of root was 5.8cm, the rooted seedlings grew very well.(5) Shoots could be regenerated from various parts of plantlet, but the bulb basal plate was higher than other parts and the initiation rate was 92.74%.(6) Transplanting: Transplanting substrate with 1/5sawdust, 1/5perlite, and 3/5humus was the best, where the survival rate could reach 80%, and it's best to transplant the plantlets in Spring, in which the survival rate could be 83.72%.
