Objective:To observe the effect of Qiqi Guben (QQGB) decoction on the levels of IL-1and TNF-a in the serum of rats with ulcerative colitis (UC), as well as the changes in the colonic mucosas of rats with UC by visual inspection, discuss the mechanism by which QQGB decoction plays a role in UC and provide basis for the traditional Chinese medicine treatment of UC patients.Methods:40SD rats were randomly divided into normal group, model group, sulfasalazine (SASP) group (western medicine group) and QQGB decoction group (TCM group),10rats for each group. A UC rat model was established by means of regional infusion with trinitrobenzene sulfonic acid (TNBS) solution. After successful modeling, the TCM group was administrated with QQGB decoction by gastric perfusion, the western medicine group with SASP solution, and the normal group and the model group with normal saline once per day for14consecutive days. After the end of the treatment, all animals were kept from eating (except water) for24hours. For each rat,2ml of blood was taken from the abdominal aorta and injected into a test tube. All rats were killed by cervical dislocation and fixed on the dissecting table before removing their colon tissues. The changes in the colon mucosas of rats were observed with the naked eye. Then, the pathomorphological changes in the colon tissues of rats were examined under light microscope. The levels of IL-1and TNF-a in the intestinal serum of UC rats were detected by ELISA test.Results:1.The general conditions of the rats in the QQGB decoction group and the SASP group were much better than before as compared with the model group:their bloody purulent and mucous stools disappeared and normal stools were formed gradually, with their spirit, appetite, mobility and flexibility regained.2. The pathomorphological changes in the colon tissues of rats were observed by visual inspection:For rats in the QQGB decoction (TCM) group and the SASP (western medicine) group, their colon tissue lesions were alleviated, mucosa congestion and edema were relieved and disappeared, and mucosa congestion and edema largely disappeared compared with the model group, while the number of mucosa congestions, edemas, ulcers or erosions as well as viscous resistance in the surrounding tissues of rats in the TCM group were smaller than the western medicine group. According to the scoring criteria for colonic mucosa pathological change of UC rats, the scores of all groups were observed:there was an extremely distinct difference between the TCM group and the model group (P<0.01), but there was no statistically significant difference between the TCM group and the model group (P>0.05).3. The pathomorphological changes in the colon tissues of rats were observed under light microscope: Compared with the model group, the colonic strucutre of rats in the QQGB decoction group and the SASP group almost entirely restored to normal and integrity. However, a small amount of inflammatory cellular infiltration remained visible, and there was a noticeable non-continuous mucous layer around the ulcer. The mucous epithelium was made up of goblet cells, the pancreatic structures were complete, and submucosa, muscularis and serosa demonstrated clear structures.4. The contents of IL-1and TNF-a in the serum of rats in the normal group were significantly decreased compared to the model group (P<0.01), those in the QQGB decoction group and the SASP group were significantly decreased compared to the model group (P<0.01), and those in the QQGB decoction group were significantly decreased compared to the SASP group (P<0.05).Conclusions:1. The occurrence and development of UC are associated with IL-1and TNF-α, and QQGB decoction can reduce their levels in serum, reduce inflammatory responses in the intestine and help improve the illness.2. Both QQGB decoction and SASP can has some therapeutic effects on UC, obviously improve the pathological change of the colon tissue and repair affected intestinal walls. However, QQGB decoction has a better curative effec than SASP.
