Mushroom polysaccharide is an important bio-active polymer material. It has anti-virus, anti-tumor immune function and so on. Mushroom resources are abundant in China and provide the basis for study of mushroom polysaccharide extract. However, the polysaccharides of Pleurotus reported less, it's has some medicinal value and it can reach the level of large-scale artificial cultivation. So, it has meaning and value. In addition, the extraction of the polysaccharide would produce a lot of residuals which usually are rich in water and nutrient. The residuals can be used as protein feed to solve a shortage of protein in livestock industries. In this study, Pleurotus as experimental material, from the choice of methods from the polysaccharide, polysaccharide extraction process optimization and the residue resource utilization three aspects to research.First, use the single factor test, compare the extraction of polysaccharides from water, formulation and enzyme. The results show that Based on water extraction, the polysaccharide extraction approached optimization at the condition of 90℃, 20:1 of ratio of liquid to solid, 3h extraction and 3 fold of ethanol dosage. By enzyme extraction, the optimal condition was 90℃, 20:1 of ratio of liquid to solid, 2h extraction and 2 fold of ethanol dosage. In addition, based on alkali extraction, the optimal condition was 50℃, 20:1 of ratio of liquid to solid, 1.5h extraction and 2 fold of ethanol dosage. Base reference time-consuming, less energy consumption, extraction rate, to practical use to which industrial production.Second, by quadratic regression orthogonal rotation test, finalized that the polysaccharide extraction rate optimal in the condition of the ethanol dosage within 3.34 ~ 4.0 times and the ratio of liquid to solid within 38.3 ~ 43.0:1, the extraction time within 3.4 ~ 3.9h and the temperature within 95.1 ~ 97.5℃. In this context, Optimum condition was temperature 90℃, extraction time 3h, liquid to solid ratio 30:1, 3 times the amount of ethanol.Finally, the orthogonal design, we researched the solid fermentation for the residuals produced in polysaccharide extraction. The residuals can be as ingredient addition in protein feed. Solid fermentation can improve the nutritional value of bacterial residuals. Optimal conditions for fermentation are 8 : 2 of residual : wheat bran, 13% yeast, 96h of culture at 30 ℃.In summary, the use of alkali formulation of the extraction of polysaccharides from Pleurotus optimized, effectively increase the extraction rate of polysaccharides. Also shown that residue as the feasibility of protein feed and protein feed materials for the search for a new way.
